基于神经元凋亡和自噬交互作用研究岭南头皮针对脑外伤昏迷大鼠的促醒作用
Lingnan Scalp Needle Promoting Awakening in Brain Traumatic Coma Rats by Regulating the Interaction of Apoptosis and Autophagy

Abstract
Objective:To investigate the mechanism of Lingnan scalp needle to promote the awakening of comatose rats with traumatic brain injury through the interaction of autophagy-related proteins(Beclin-1,p-62,LC3B)-apoptosis-related proteins(Cleaved-Caspase3,Bcl-2)signaling axis.Methods:Forty rats were randomly divided into control group,model group,sham needle group and Lingnan scalp needle group according to body weight,with 10 rats in each group.The rat model of traumatic coma(TBI)was constructed by free-fall impingement method,TBI+Lingnan scalp needle method was used in the Lingnan scalp needle group,TBI+ordinary acupuncture method was used in the sham needle group,the modified neu-rological severity score(mNSS)were compared in each group,the histopathological changes of cortex and hippocampus were observed by HE stain-ing,and the autophagy proteins[LC3B,p-62,Beclin-1]and apoptotic proteins[Caspase-3,Bcl-2]were detected by Western blot method Level change.Results:On days 0,7 and 14,the mNSS of the TBI group,sham needle group and Lingnan scalp needle group were higher than those of the control group(P<0.01);on days 7 and 14,the mNSS of the sham needle group and Lingnan scalp needle group were lower than those of the TBI group(P-<0.0 I),and the mNSS of the rats in the Lingnan scalp needle group were lower than those of the sham needle group(P<0.01),and on day 14,the mNSS of the rats in the Lingnan scalp needle group were lower than the mNSS on day 7(P<0.01).After Lingnan scalp needle treatment,the number of cortical neurons in the Lingnan scalp needle group was abundant,locally arranged irregularly,and a small number of neuronal nuclei were seen in the cortex wrinkled and deeply stained,with unclear demarcation of nucleus and cytoplasm,and the morphology of neuronal cells in the hip-pocampus was regular,arranged neatly and densely packed with a large,rounded nucleus that was uniformly light blue or blue in color,with a clear nucleolus and abundant cytoplasm.Compared with the control group,the expression of apoptotic protein Cleaved-Caspase3 was elevated(P<0.05),the expression of Bcl-2 protein was decreased(P<0.05),the expression of autophagy protein LC3B was increased(P<0.05),the expression of Beclin-1 was elevated(P<0.01),and the expression of p-62 was decreased(P<0.05)in the TBI group;and compared with the TBI group,sham needle group,apoptotic protein Bcl-2 protein expression was elevated(P<0.05),autophagy protein LC3B expression was decreased(P<0.01),and the difference in the expression of p-62,Beclin-1,and Cleaved-Caspase3 between the two groups was not statistically significant(P>0.05),and in the Lingnan scalp needle group,apoptotic protein Bcl-2 protein expression was elevated(P<0.0l),autophagy protein LC3B,Beclin-1 expression decreased(P<0.01),the difference in p-62,Cleaved-Caspase3 expression between the two groups was not statistically significant(P>0.05).Conculusion:Lingnan scalp needle can effectively promote the recovery of coma rats after craniocerebral injury,and its mechanism may be related to the inhibition of the expres-sion of autophagy and apoptotic proteins.关键词
