摘要

Abstract

[Objective]In order to select the culture to purificate and preservate of wild Lentinula edodes,to provide suitable conditions for cultivating and preserving germplasm resources of wild Shiitake strains.[Method]Cultured from strain comes to spawn of Shiitake which collected from Antu County,Jilin Province,set up four kinds of purification and preservation medium,compared the difference of growth of mycelium and the colorof the colonies.[Result]Both hyphal may germinate in different purificated and preserve cultures of the spawn of L.edodes,but has big differences.The results showed that the hyphal of L.edodes could grow in all 4 culture,and formula 4(patato 200 g,sawdust 10 g,soybean powder 10 g,glucose 20 g,agar 20 g,water 1 L)was much more suitable for the growth of this strain,which was better than other culture conditions.And the growth rate of mycelium was the highest,reaching 0.51 cm/d,vigorous,white,and the colonies without pigment.Formula A(wood chips 78%,wheat bran 20%,lime 1%,gypsum 1%)was more suitable for long-term culture and preservation of mother spawn,and the germination rate of mycelium is higher,and its growth is vigorous and neat,and the colonies without pigment,may preserve 15 months.[Conclusion]Studies on selecting the cultures for purification and preservation of wild L.edodes,which laid a foundation for the collection,purification and long-term preservation of wild resources of L.edodes.
