基于UPLC-MS/MS羊肚菌子实体生物碱类代谢物差异分析OACSTPCD

Differential Analysis of Alkaloid Metabolites in Morchella Fruiting Body by Using UPLC-MS/MS

为了解羊肚菌Morchella菌株(MLM、MTL)子实体生物碱类化合物,同时比较其中生物碱类化合物差异,利用超高效液相色谱串联质谱(UPLC-MS/MS)、聚类分析、样品相关性分析和正交偏最小二乘判别等分析方法,在羊肚菌菌株MLM和MTL的子实体中共检测到78种生物碱代谢物(其中菌株MLM子实体77种生物碱代谢物,菌株MTL子实体75种生物碱代谢物),包括生物碱(34种)、吡啶类生物碱(6种)、吡咯类生物碱(2种)、酚胺(13种)、喹啉类生物碱(2种)、哌啶类生物碱(2种)、异喹啉类生物碱(1种)、吲哚类生物碱(18种);在羊肚菌菌株MLM和MTL的子实体中共筛选得到差异代谢物43种,相对于菌株MTL,MLM有8种成分上调;相对于菌株MLM,MTL有35种成分下调,其中甜菜碱在MLM中含量最高,N-(2-羟基-4-甲氧基苯基)乙酰胺在MTL中含量最高;N-对香豆酰羟基腐胺、对香豆酰胆碱、N-阿魏酰胍丁胺仅在MTL中检测到,而咖啡酰胍丁胺只在MLM含有;在显著差异代谢产物中MTL中水杨酰胺含量最高,而在MLM中3-羟基-2-氨基苯甲酸含量最高,说明MLM比MTL有更高的营养保健价值.差异代谢物主要分布在19种代谢途径中,包括新陈代谢途径通路、次生代谢的生物合成通路、色氨酸代谢通路、精氨酸和脯氨酸代谢通路以及赖氨酸降解等代谢通路.通过分析2种羊肚菌子实体中生物碱差异代谢物的相关生物学特性和代谢途径,为日后羊肚菌在保健食品、化妆品、药品等领域的应用提供参考依据.

In order to investigate and compare the alkaloid compositions between fruiting bodies cultivated with different strains of Morchella,MLM and MTL,methods of UPLC-MS/MS,cluster analysis,sample correlation analysis and orthogonal partial least squares-discriminant analysis were used at the present study.A total of 78 alkaloid metabolites were detected in the fruiting bodies from the MLM and MTL strains of Morchella(77 from MLM and 75 from MTL),which included 34 kinds of alkaloids,six kinds of pyridine alkaloids,two kinds of pyrrole alkaloids,13 kinds of phenolamines,two kinds of quinoline alkaloids,two kinds of piperidine alkaloids,one kind of isoquinoline alkaloids,and 18 kinds of indole alkaloids.A total of 43 metabolites were successfully screened to show a differential change of content in the fruiting bodies between MLM and MTL,in which eight metabolites were upregulated in MLM as compared to MTL.Compared with the MLM,on the other hand,35 metabolites of MTL were downregulated.In MLM,betaine had the highest content but in MTL it was N-(2-hydroxy-4-methoxy phenyl)acetamide.N-p-coumaryl hydroxy-putcine,p-coumaryl choline and n-feruloguidine were found only in MTL while caffeylguidine existed only in MLM.Among the metabolites showing differential changes with a significant difference,salicylamide and 3-hydroxy-2-aminobenzoic acid revealed the highest content in MTL and MLM,respectively,indicating that MLM might have a higher value for nutrition supply and health-care than MTL.Metabolites showing the differential changes between the two strains were mainly distributed within 19 metabolic pathways,including those of biosynthesis pathway of secondary metabolism,tryptophan metabolic pathway,arginine and proline metabolic pathway,and lysine degradation metabolic pathway.The biological characteristics and metabolic pathways analyzed from the alkaloids with differential changes in the fruiting bodies of two different Morchella strains(MLM and MTL)at this study should provide a reference for further development of edible fungi carrying various functions in Morchella.
