摘要

Abstract

Objective To investigate the effect of the fibroblast growth factor 2(FGF2)on the development of in vitro cultured kidney tissues from the 12.5-day embryonic mice,and explore its relationship with the development of renal corpuscle.Methods The kidney tissues of 12.5-day embryonic mice were in vitro cultured with 0,25,100,200,400 ng/mL FGF2 for 2,4,6 d,and then the develop-ment of renal corpuscles was observed by HE staining.Immunohistochemical staining combined with stereo-logical methods were used to observe and semi-quantitatively analyze the expression of FGFR1 in renal corpuscles at different developmental stages.Results The development process of kidney tissues in vitro was similar to the development process in vivo.After cultured with FGF2,the number of renal corpuscles at different stages of development process increased.Immunohistochemical staining and stereo-logical results showed the expression of FGFR1 in renal tissue in vitro cultured with 25,100,200,400 ng/mL FGF2 was significantly higher than that of 0 ng/mL FGF2(P<0.05),but there was no statistically significant difference among four concentration groups.Conclusion FGF2 may promote the aggregation of mesenchymal cells and the development of renal corpuscles by binding to FGFR1 during the kidney develop-ment process.
