Abstract

With the rapid development of the aquaculture industry,the aquaculture water environment is deteriora-ting day by day,and the diseases of farmed animals are frequent.Aquatic probiotics are widely used in the im-provement of aquaculture water environment because of their environmental friendliness and safety.However,bac-teria are susceptible to the external environment and have low stability,which seriously restricts their application in the aquaculture water environment.The immobilized microbial technology can greatly improve the stability of domi-nant bacteria in the water environment,which provides a direction for the application of bacteria in water purifica-tion.In this study,a highly nitrite-degrading strain X1 was screened from the rhizosphere soil of mangrove forest in Egret Park,Sanya,and the nitrite degradation rate of this strain could reach 97.18% on the 4th day.Phylogenetic tree analysis of 16S rRNA gene showed that the strain belonged to Pseudomonas stutzeri.The strain was combined with sodium alginate-chitosan composite carrier to form immobilized pellets,which were put into aquaculture wastewater,and the nitrite degradation rate could reach 98.15% on the 2nd day,and the degradation rate on the 3rd and 4th days remained stable,and there was no nitrite accumulation,reaching 98.35% on the 5th day.The results confirmed that Pseudomonas stutzeri schudei could degrade nitrite in water efficiently,quickly and safely,and the immobilization technology could better improve the degradation rate and stability of the strain,which pro-vided a certain reference for the application of immobilized Pseudomonas stutzeri in water purification.
