Expression and Clinical Value of CILP2 in Endometrial Cancer

Abstract
Objective:To validate the expression and clinical significance of CILP2 gene in endometrial carcinoma through tissue-level experiments by utilizing an online bioinformatics database,and investigate the relations of CILP2 gene expression to prognosis and immune infiltration.Meth-ods:The expression of CILP2 in pancarcinoma was obtained from the TIMER database and validated using the DriverDBv3 database.Differential expression analysis of 552 cases of endometrial cancer and 35 cases of normal endometrial tissue data downloaded from the TCGA database was performed using R software.The Human Protein Atlas database was utilized to ana-lyze the positive expression of CILP2 in endometrial cancer.Clinical correlation analysis on the endometrial cancer data downloaded from TCGA database was conducted using R software,and the expression of CILP2 in endometrial cancer tissue chips was further detected by immunohistochemical method.Mutation information of the CILP2 gene was obtained from cBio-Portal database.GEPIA,UALCAN and Linkedomics databases were used to analyze survival time for high and low expression groups of CILP2.Univariate and multivariate Cox analyses as well as diagnostic ROC analysis results were obtained using R software.A protein network interacting with CILP2 was constructed using STRING database;co-expressed genes of CILP2 were retrieved from Linkedomics database;these co-expressed genes were analyzed and visualized by GO and KEGG path-ways using R software;finally,the relations of CILP2 to tumor-infiltrating immune cells and immune regulatory factors were analyzed using TISIDB.Results:The mRNA and protein levels of CILP2 gene in endometrial carcinoma tissue were signifi-cantly higher than those in normal tissue.Immunohistological results confirmed the abnormally high expression of CILP2 in endometrial carcinoma.cBioPortal mutation analysis showed that CILP2 was highly expressed in the altered group,and the survival time of endometrial cancer patients in the variant group was significantly lower than that in the unaltered group.Sur-vival analyses of GEPIA,UALCAN and Linkedomics also found that the survival period of patients with high CILP2 expres-sion was significantly shorter than that of patients with low CILP2 expression.Meanwhile,univariate and multivariate Cox an-alyses results showed that high CILP2 expression was associated with poor prognosis of patients.The results of STRING on-line analysis showed that the proteins interacting with CILP2 were involved in biological processes including transcriptional regulation,lipid metabolism,ubiquitin protein modification and immune cell differentiation.GO and KEGG enrichment ana-lyses showed that CILP2 co-expressed genes in endometrial carcinoma tissue might be involved in the positive regulation of ubiquitin protein transferase activity,glycosylation,protein metabolism,exocytosis vesicles,ribonucleoprotein complex bind-ing,and related to signaling pathways such as glycosylation,fatty acid degradation,cell cycle and insulin secretion.TISIDB analysis showed that CILP2 was negatively correlated to activated CD8+T cells,macrophages and other tumor-infiltrating cells,and was positively correlated to some immune-stimulating factors such as CD40,CD80 and IL6;CILP2 was positively correlated to some immunosuppressive factors such as PVRL2 and TGFB1;and CILP2 was negatively correlated to major his-tocompatibility complex factors such as HLA-DMA and HLA-DOB.Conclusion:Based on bioinformatics and tissue-level ex-periments,it has been revealed that CILP2 is aberrantly highly expressed in endometrial cancer tissue,and its expression level is closely related to the poor prognosis and immune infiltration of endometrial cancer patients.CILP2 gene may be a mo-lecular marker for assessing patient prognosis and a target for gene therapy.关键词
