Associations of cheese intake with osteoarthritis:a Mendelian randomization study

目的:采用孟德尔随机化的方法,探讨奶酪摄入量与骨关节炎发生风险的关联性.方法:采用全基因组关联分析数据进行研究,暴露因素为奶酪摄入量,结局变量为骨关节炎.采用逆方差加权法(IVW)法、加权中位数法、MR-Egger法进行孟德尔随机化分析,并进行多效性分析、异质性检验、敏感性分析.结果:IVW法和加权中位数法结果显示,奶酪摄入量为骨关节炎发病的保护因素,二者之间存在负向因果关联(P<0.05).MR-Egger回归结果显示,奶酪摄入量与骨关节炎发病关联无统计学意义(P=0.335).研究结果经检验不存在异质性和多效性,敏感性分析未发现导致总体因果关系发生显著变化的单核苷酸多态性位点.结论:奶酪摄入量与骨关节炎发生风险存在因果关系.

Objective:Using Mendelian randomization method,explore the causal relationship between cheese intake and the risk of osteoarthritis.Methods:Data from gene-wide association studies were used for analysis.The exposure factor was cheese intake,and the outcome variable was osteoarthritis.Mendelian randomization(MR)analysis was performed by IVW method,MR-Egger method and weighted median,and pleiotropy test,heterogeneity test and sensitivity analysis were performed.Results:The results of IVW and weighted median showed that cheese intake was a protective factor for osteoarthritis,and there was a negative causal association between them(P<0.05).The MR-Egger regression showed no statistically significant association between cheese intake and osteoarthritis(P=0.335).The results were tested for no heterogeneity or pleiotropy,and sensitivity analyses did not identify SNPs that significantly changed the overall causal relationship.Conclusion:Cheese intake is causally related to osteoarthritis.
