目的:研究不同配伍比例的半枝莲-白花蛇舌草药对(简称半白药对,SB-OD)对肝癌细胞株(MHCC97H细胞和Huh7 细胞)增殖活性的抑制作用.方法:运用CCK-8 法观察不同配伍比例的半白药对(半白1∶1,半白2∶1,半白1∶2)干预MHCC97H和Huh7 细胞24 h,分别计算其IC50值;并应用细胞平板克隆形成实验检测细胞克隆形成能力,分析不同配伍比例的半白药对对MHCC97H和Huh7 细胞增殖活性的影响.结果:不同配伍比例的半白药对作用MHCC97H和Huh7 细胞后的生存活力明显下降(P<0.01),并呈浓度依赖性,其中半白 1∶1 组作用于 MHCC97H 和 Huh7 细胞的 IC50 值分别为6.69 mg/mL和7.95 mg/mL;半白2∶1 组作用于MHCC97H和Huh7 细胞的IC50值分别为2.58 mg/mL和3.43 mg/mL;半白 1∶2 组作用于 MHCC97H 和 Huh7 细胞的 IC50 值分别为 15.04 mg/mL 和16.60 mg/mL.不同配伍比例的半白药对干预MHCC97H和Huh7 细胞后的克隆形成率明显降低(P<0.01).结论:不同配伍比例的半白药对均能够抑制肝癌MHCC97H和Huh7 细胞的增殖活性,2∶1 配伍比例的抑制作用最佳,可作为半白药对抗肿瘤研究的实验基础.

Objective:To study the inhibitory effect of Scutellariae Barbatae Herba(SBH)and Hedyotis Diffusae Herba(HDH)couplet intervention with different compatibility ratio on the proliferation activity of liver cancer cell lines(MHCC97H cells and Huh7 cells).Methods:CCK-8 assay was used to observe the intervention of MHCC97H and Huh7 cells with different compatibility ratio of SBH and HDH(1∶1,2∶1,1∶2)for 24 h,and their IC50 values were calculated respectively.The cell plate clone formation experiment was used to detect the cell clone formation ability.The effects of different compatibility ratios of SBH and HDH on the proliferation activity of MHCC97H and Huh7 cells were analyzed.Results:The survival activity of MHCC97H and Huh7 cells significantly decreased(P<0.01)with different compatibility ratios of SBH and HDH in a concentration-dependent manner.The IC50 values of group 1∶1 were 6.69 mg/mL and 7.95 mg/mL,respectively;the IC50 values of group 2∶1 were 2.58 mg/mL and 3.43 mg/mL,respectively;and the IC50 values of group 1∶2 were 15.04 mg/mL and 16.60 mg/mL,respectively.The clone formation rate of MHCC97H and Huh7 cells was significantly reduced after intervention with different compatibility ratios of SBH and HDH(P<0.01).Different compatibility ratios of SBH and HDH have a significant inhibitory effect on the proliferation activity of MHCC97H and Huh7 cells.Conclusion:Different compatibility ratios of SBH and HDH can all inhibit the proliferation activity of liver cancer MHCC97H and Huh7 cells,and the best inhibition effect is achieved with a 2∶1 compatibility ratio,which can serve as an experimental basis for SBH and HDH couplet anti-tumor research.
