沼泽红假单胞菌CGA009砷调控蛋白(ArsR)的调控作用OACSTPCD
Regulatory Role of Arsenic Regulatory Protein(ArsR)in Rhodopseudomonas palustris CGA009
采用改良的凝胶电泳迁移滞缓(EMSA)法和砷生物传感法研究沼泽红假单胞菌CGA009菌株6个高度异质性ArsR相关蛋白的调控作用.研究结果表明:6个ArsR相关蛋白都具有As(Ⅲ)依赖的调控活性,改良的EMSA图谱可用于系统评价ArsR的功能;6个相关蛋白对异种(E.coli pR733 or Stapylococcus aureus pⅠ 258)的砷抗性操纵子的调控具有不同的选择性,氨基酸序列同源性(甚至低于30%同源性)的ArsR具有砷依赖的调控作用.
Using a modified gel electrophoretic mobility shift assay(EMSA)method and Arsenic biosensing method,the regulation of six highly heterogeneous ArsR-associated proteins from Rhodopseudomonas palustris CGA009 bacterial strain is investigated.The reseach results show that six ArsR-associated proteins exhibited As(Ⅲ)-dependent regulatory activity.The modified EMSA mappings can systematically evaluate the functions of ArsR.Six ArsR-associated proteins have different selectivity in regulating the arsenic resistance o-peron of different species(E.coli pR733 or Staphylococcus aureus p Ⅰ 258).ArsRs with amino acid sequence homology(even with below 30% homology)have arsenic-dependent regulation.
