Effect of Exendin-4 on the spontaneous discharge of neurons in the substantia nigra pars reticulata of mice

Abstract
Objective To investigate the effectof Exendin-4,a glucagon-like peptide-1(GLP-1)receptor agonist,on the spontaneous discharge of neurons in the substantia nigra pars reticulate of mice.Methods Immunofluorescent staining was used to observe the expression of GLP-1receptor(GLP-1R)in the substantia nigra pars reticulata of mice.Extendin-4 10 μmol/L were injected onto the surface of neurons in the substantia nigra pars reticulate using in vivo electrophysiological microelectrodes to ob-serve the spontaneous discharge of non-dopaminergic neurons(assumed to be GABAergic neurons)in the substantia nigra pars re-ticulate.Results Immunofluorescent staining showed a high expression level of GLP-1R in the substantia nigra par reticulate of mice.As for the 13 neurons recorded in the substantia nigra par reticulata,Exendin-4 significantly reduced the spontaneous dis-charge rate of 9 neurons,with a significant reduction in discharge frequency from(18.62±3.19)Hz to(14.615±3.06)Hz(t=7.257,P<0.05),and the discharge frequency was reduced by(21.85±7.72)%on average,which was significantly higher than the reduction rate of(1.15±9.67)%in the normal saline group(Z=2.282,P<0.05).In the other 4 neurons,Exendin-4 had no signi-ficant effect on discharge frequency,which changed from(13.49±7.16)Hz to(13.54±7.45)Hz.Conclusion Exendin-4 can re-duce the spontaneous discharge frequency of some neurons in the substantia nigra pars reticulata.关键词
