毛头鬼伞漆酶基因Cc_lac21的克隆及生物信息学和组织表达分析OA北大核心CSTPCD
Cloning,Bioinformatics and Expression Analyses of Laccase Gene Cc_lac21 in Coprinus comatus
为了确定毛头鬼伞基因组中漆酶基因Cc_lac21的"缺口"序列及可能的生理功能,本研究对其进行克隆和生物信息学分析,并以18S rDNA为内参基因,检测菌丝阶段以及商品成熟阶段子实体的菌盖、菌柄顶端和菌柄基部Cc_lac21的表达水平.结果表明,Cc_lac21基因全长2 243 bp,含有12个内含子、13个外显子,编码序列为1 452 bp,编码483个氨基酸.Cc_Lac21为稳定的亲水性蛋白,N端前17个氨基酸为信号肽序列,分泌到胞外;二级结构以无规则卷曲和延伸链为主;在28个丝氨酸(S)、21个苏氨酸(T)和5个酪氨酸(Y)上可能发生磷酸化修饰;在第286位的苏氨酸和第381位的丝氨酸处可能发生O-糖基化修饰;含有3个铜氧化蛋白结构域,分别是2个铜氧化还原蛋白超家族(cupredoxin super family)和1个云芝Tv-LCC样真菌漆酶的第三个铜氧化还原域(CuRO_3_Tv-LCC9);与鳞白环蘑漆酶的亲缘关系最近,包含3个基序.通过RT-qPCR分析发现,Cc_lac21 在菌丝体阶段和成熟菌柄基部表达量高,在菌盖和伸长生长的菌柄顶端表达量低,推测Cc_lac21可能参与基质木质素的降解以及菌柄中菌丝细胞壁的成熟过程.本研究结果可为探究漆酶基因参与菌柄细胞壁成熟机制奠定基础.
To determine the'gap'sequence and possible physiological functions of the laccase gene Cc_lac21 in the genome of Coprinus comatus,the Cc_lac21 gene was cloned and analyzed by bioinformatics,and its expression level at mycelium stage and in the pileus,upper stipe and basal stipe of fruiting bodies at commercial maturation stage were detected using 18S rDNA as reference gene.The results showed that the full length of Cc_lac21 was 2 243 bp containing 12 introns and 13 exons,and the coding sequence was 1 452 bp encoding 483 amino acids.Cc_Lac21 was a stable hydrophilic protein with the first 17 amino acids at N-termi-nus as signal peptide sequence,which was secreted to extracellular space.The secondary structure of Cc_Lac21 was mainly composed of random coiled-coil and extended chain.Phosphorylation might occur on 28 serines(S),21 threonines(T)and 5 tyrosines(Y),and O-glycosylation might occur at threonine 286 and serine 381.It contained three cupredoxin protein domains including two cupredoxin super families and one CuRO_3_Tv-LCC9 of Trametes versicolor Tv-LCC like fungal laccase.It was the most closely related to laccase of Leucoagaricus leucothites and contained three motifs.Through RT-qPCR analysis,it was found that Cc_lac21 was highly expressed in the mycelium and basal stipe of matural fruiting bodies,and was lower expressed in the pileus and upper stipe in elongation growth,speculating that Cc_lac21 might participate in the lignin degradation of cultivated substrate and maturation process of stipe cell wall.The results of this study could lay a foundation for exploring the mechanism of laccase genes involved in cell wall maturation of stipe.
