重楼皂苷Ⅰ通过调控Notch信号通路对前列腺癌PC-3细胞迁移侵袭力的影响
Effect of Polyphyllin Ⅰ on the Migration and Invasion of Prostate Cancer PC-3 Cell Migration by Regulating Notch Signaling Pathway

Abstract
Objective To observe the effect of Polyphyllin Ⅰ on the migration and invasion of prostate cancer PC-3 cells and explore its mechanism.Methods Prostate cancer PC-3 cells were cultured in vitro and divided into model group,positive control group(cisplatin 2.5 µg/mL),polyphyllin Ⅰ low,medium,and high dose group(0.25,0.5,1.0 µg/mL).Real time quantitative PCR was used to detect the mRNA expression of Notch1,Jagged1,and hairy and enhance-of-split homologues 5(Hes5).Western Blot was used to detect the protein expression of Notch1,Jagged1,and Hes5.Cell scratch assay and Transwell assay were used to detect changes in PC-3 cell migration and invasiveness.Transmission electron microscopy was used to observe the morphological changes of pseudopodia in PC-3 cells.Results Many pseudopodial like protrusions were observed on the surface of PC-3 cells in the model group.Compared with the model group and positive control group,the number of pseudopodial like protrusions on the surface of PC-3 cells in polyphyllin Ⅰ low,medium,and high dose group decreased.Compared with the model group,the mRNA and protein expression of Notch1,Jagged1,and Hes5,as well as the wound healing rate and invasive cell count in the positive control group and polyphyllin Ⅰ low,medium and high dose groups reduced(P<0.01).Compared with the positive control group,the mRNA and protein expression of Notch1,Jagged1,and Hes5,as well as number of invasive cells in polyphyllin Ⅰ medium and high dose group reduced(P<0.01).Compared with the positive control group,the wound healing rate in polyphyllin Ⅰ low,medium and high dose group reduced(P<0.05,P<0.01).Conclusion Polyphyllin Ⅰ could inhibit the migration and invasion of prostate cancer PC-3 cells,and its mechanism might be related to downregulation of key proteins in the Notch signaling pathway of PC-3 cells and inhibition of pseudopodia formation on the surface of PC-3 cells.关键词
