贝莱斯芽孢杆菌(Bacillus velezensis)SY01对苹果树腐烂病菌(Valsa mail var.pyri)SW5具有较强的抑制作用.为探明其对苹果树腐烂病的作用机理,本研究采用荧光定量 PCR 检测菌株SY01对苹果树抗性关键基因表达的影响.并通过脂肽类抑菌成分合成基因的扩增比对与UPLC-Triple TOF-MS/MS相结合的方法初步分析菌株SY01中脂肽类抑菌物质成分.结果表明,在菌株SY01诱导下,苹果树的5种关键抗性相关基因的表达峰值时间存在差异,其中MdNPR1、MdPDF1、MdPR2、MdPR4分别在24、72、72和48 h达到峰值,而MdPR1在0~96 h之间呈现持续上调的趋势.菌株SY01的抗菌脂肽粗提物在20~40℃和pH为7的条件下具有较强的抑菌活性,并对自然光和紫外线照射有较好的适应能力.脂肽类抑菌物质合成基因的扩增比对分析结合UPLC-Triple TOF-MS/MS的结果表明,菌株SY01可通过产生Iturin、Fengycin、Surfactin等抗菌次级代谢产物来抑制菌株SW5的生长.综上所述,菌株SY01能够通过诱导苹果树关键抗性基因的表达,以及分泌抗菌次级代谢产物来抑制苹果树腐烂病菌的生长,从而实现对苹果树腐烂病的防治.

Bacillus velezensis SY01 has a strong inhibitory effect on the apple tree canker pathogen(Valsa mail var.pyri).In order to explore its prevention and control mechanism against apple tree canker,quantitative PCR was used to detect the effect of strain SY01 on the expression of key genes for apple tree canker disease resistance and to determine the composition of lipopeptide substances in strain SY01 by combining the expression difference analysis of the lipopeptide synthesis genes and UPLC-Triple TOF-M S/MS.The results showed that there were differences in the expression peak time of five key resistance-related genes in apple trees induced by strain SY01.MdNPR1,MdPDF1,MdPR2,and MdPR4 reached their peak at 24,72,72 and 48 h,respectively,while MdPR1 showed a sustained upward trend at 0-96 h.The crude extracts of antimicrobial lipopeptides of strain SY01 showed strong antibacterial activity under conditions of 20-40℃,natural light irradiation,ultraviolet irradiation,and pH 7 and had a good adaptability to natural light irradiation and UV irradiation.The amplification comparison analysis of lipopeptide inhibitory substance synthesis genes combined with UPLC-Triple TOF-MS/MS showed that strain SY01 could inhibit the growth of strain SW5 through the production of antimicrobial secondary metabolites such as Iturin,Fengycin and Surfactin.In summary,strain SY01 can inhibit the growth of apple tree canker fungus by inducing the expression of key resistance genes and secreting antimicrobial secondary metabolites for the control of apple tree canker disease.
