摘要

Abstract

Objective:To treat rabbit knee osteoarthritis(KOA)by needle knife"unknotting-method",and to investigate its effect on PI3K/AKT/mTOR related factors in femoral condyle cartilage tissue.Methods:A total of 24 SPF male New Zealand white rabbits were randomly divided into blank group,model group and needle knife group,8 rabbits/group.The model group and the needle knife group were prepared by Videman-modified-method.After successful modeling,the needle knife group were treated with the needle knife"unknotting-method"to loosen the tendon lesions around the knee on the affected side,and the blank group and the model group were equally grasped and fixed for 5 mins,without any intervention,once a week,4 times in a row.One week after the last intervention,HE staining and Mankin's score were used to observe the pathological morpho-logical changes of femoral condyle cartilage tissue of the affected knee joint in the three groups,the mRNA transcription levels of PI3K,AKT and mTOR in the femoral condyle cartilage tissue of the affected knee joint in the three groups were detected by qPCR,and the protein expression levels of PI3K,AKT and mTOR in the femoral condyle cartilage tissue of the affected knee joint in the three groups were detected by Western-blot.Results:Compared with the blank group,the HE staining results showed that the surface layer of femoral condyle cartilage tissue of the affected knee joint in the model group was missing and uneven,the cluster and arrangement of transitional cells were irregular,the Mankin's score was increased(P<0.05),the mRNA transcription levels and protein expression of PI3K and AKT were up-regulated(P<0.05),while the mTOR transcription level and protein expression were up-regulated(P<0.05);Compared with the model group,the results of HE staining showed that the morphology and alignment integ-rity of the cartilage tissue of the femoral condyle of the affected knee joint in the needle knife group tended to be blank group,the occasional chondro-cyte cluster,and the tidal line were more complete than that of the model group,and the Mankin's score was significantly lower(P<0.05),the mRNA transcription levels and protein expression of PI3K and AKT were down-regulated(P<0.05),and the mRNA transcription level and protein expres-sion of mTOR were up-regulated(P<0.05).Conclusion:The needle knife"unknotting-method"can improve the degeneration of femoral condylar car-tilage of the affected knee joint and effectively protect the function of the affected knee joint,and its mechanism may be related to the transcription level of PI3K/AKT/mTOR pathway-related factors.
