多花黄精Polygonatum cyrtonema根腐病是制约其产量和质量的重要因素.为探索该病害绿色有效的生物防控途径,采用组织分离法获得多花黄精根茎内生真菌菌株HJ-53和多花黄精根腐病病原真菌.结合形态学特征和ITS/TEF序列分析对真菌菌株进行鉴定,确定内生真菌HJ-53为桔青霉Penicillium citrinum;多花黄精根腐病病原为尖孢镰刀菌Fusarium oxysporum,并通过科赫法则验证.采用平皿对峙培养和生长速率法初步测定了菌株HJ-53及发酵液乙酸乙酯提取物的抑菌活性,利用单因素及正交试验筛选了该菌株最适基础培养基和最适发酵条件.结果表明,内生真菌HJ-53具有较强的抗菌活性,其两点对峙培养和五点对峙培养的抑菌率分别达到了 54.81%和61.04%;浓度为10g·L-1的菌株发酵液乙酸乙酯提取物,其抑菌率高达73.20%.该菌最佳发酵培养基为葡萄糖30g·L-1、蛋白胨15 g·L-1和氯化钙1.5 g·L-1;最适发酵条件为发酵5 d,发酵温度25 ℃,摇床转速180 r·min-1,pH值为7.研究表明,内生真菌菌株HJ-53具有一定的生防潜力,可为多花黄精根腐病的生物防治菌剂的研究提供理论基础和菌种资源.

The root rot of Polygonatum cyrtonema is a major risk factor that restricts its yield and quality.In order to explore a green and effective biological control pathway for this disease,this study used tissue isolation method to obtain the endophytic fungal strain HJ-53 and the pathogenic fungi of P.cyrtonema root rot.Based on morphological characteristics and ITS/TEF sequence analysis,the fungal strain HJ-53 was identified as Penicil-lium citrinum;the pathogen of the root rot of P.cyrtonema was Fusarium oxysporum,which had been validated by the Koch rule.The antifungal activity of strain HJ-53 and the ethyl acetate extract from the fermentation broth were preliminarily determined using plate confrontation cultivation and growth rate methods.The optimal basic culture medium and fermentation conditions for the strain were screened using single factor and orthogonal ex-periments.The results showed that the endophytic fungus HJ-53 had strong antifungal activity,and its two points and five points confrontation cultivation had antifungal rates of 54.81%and 61.04%,respectively;the ethyl ace-tate extracted from the fermentation broth of the strain with a concentration of 10 g L-1 had a high antifungal rate of 73.20%.The optimal fermentation medium for this endophytic fungus was glucose 30 g·L-1,peptone 15 g·L-1,and calcium chloride 1.5 g·L-1;the optimal fermentation conditions were fermentation for five days,fermentation temperature of 25 ℃,shaking bed speed of 180 r-min-1,and pH value of 7.The results showed that the endo-phytic fungal strain HJ-53 has certain biocontrol potential,which provides a theoretical basis and strain resources for the research of biocontrol agents for the root rot disease of P.cyrtonema.
