添加褪黑素对常温和冷冻保存獭兔精液的影响OACSTPCD
Effects of Supplementing Melatonin on Semen Qualities of Rex Bucks under Room Temperature and Cryopreservation Conditions
褪黑素是哺乳动物松果体分泌的一种吲哚类激素,具有较强的抗氧化能力,研究褪黑素在精液保存液中的适宜添加浓度对精液的保存、运输和人工授精具有重要意义.试验采集20只獭兔公兔的精液,在常温(20℃)和冷冻(-196℃)条件下,在精液保存液中添加不同浓度(0,0.05,0.1,0.2,0.4 mmol/L)褪黑素后,观察其在常温下各时间点(12,24,36,48,60,72,96 h)和解冻前后的精子活力、精子密度、精子畸形率.结果表明:常温下添加不同浓度的褪黑素对精子活力、精子密度和精子畸形率无显著影响;冷冻条件下添加褪黑素可显著提高精子活力(P<0.05),其中,在冷冻保存液中添加0.05 mmol/L褪黑素效果较好.
Melatonin is an indoles secreted by pineal gland of mammals,which has high antioxidant effects.The study of its best concentration in semen extenders is beneficial for semen preservation,transportation and artificial insemination.In this study,20 Rex bucks were used and their semen samples were collected.After supplementing gradient amounts of melatonin(0,0.05,0.1,0.2,0.4 mmol/L),the sperm motility,density and deformity of all semen samples were observed at different time points(12,24,36,48,60,72,96 h)at room temperature,and after freezing/thawing processes.The results showed that the supplementation of melatonin under room temperature had no significant effects on sperm quality index,while melatonin significantly improved sperm motility(P<0.05)during freezing/thawing process.The best concentration for melatonin supplementation in sperm freezing extender of Rex buck is 0.05 mmol/L.
