Abstract

Objective:The CMTM6 enzyme inhibits ferroptosis and promotes oral squamous cell carcinoma(OSCC)via a molecular mechanism produced by the oral dominant microbe Fusobacterium nucleatum(Fn).Methods:In order to de-termine the viability of human SCC-9 cells,CMTM6 expression was determined in normal and OSCC tissues and Fn[multi-plicity of infection(MOI)=100∶1]was applied to normal and OSCC tissues to determine CMTM6 expression.Transfected SCC-9 cells with si-TLR4 were infected with Fn,and Fn treatment was evaluated in SCC-9 cells.The viability of cells,colony formation,proliferation of cells,iron levels,reactive oxygen species(ROS)production,and ferroptosis were recorded.Results:CMTM6 exhibited elevated expression levels in OSCC.Fn was found to augment the viability of SCC-9 cells,leading to a sig-nificant up-regulation of CMTM6 expression.Furthermore,Fn was observed to stimulate the proliferation and colony formation of SCC-9 cells,as well as decreased intracellular ROS levels and attenuated iron deposition.Furthermore,Fn was found to modulate the expression of proteins implicated in ferroptosis,such as the decreased expression of CHAC1 and PTGS2 in SCC-9 cells,as well as the increased expression of GPX4.The inhibition of CMTM6 reversed the effects of Fn on SCC-9 cells,demonstrating statistical significance(P<0.05).Conclusion:These findings suggest that Fn may suppress ferroptosis in SCC-9 cells by upregulating CMTM6 expression and potentially contributing to the pathogenesis of OSCC.
