蒺藜苜蓿转录因子基因MtC3H的克隆、亚细胞定位及表达分析

Abstract
C3H transcription factors belong to the zinc finger protein(ZFP)family and play a crucial role in plant responses to both biotic and abiotic stresses.To investigate the potential function of the MtC3H gene in Medicago truncatula,the full-length sequence of the MtC3H gene was cloned from Medicago truncatula R108.the full length of CDS of MtC3H was 672 bp and encoded 223 amino acids.Phylogenetic tree analysis showed that MtC3H protein was closely related to Cicer arietinum and Pisum sativum.Expression pattern analysis demonstrated that MtC3H exhibited the highest expression level in the stems of Medicago truncatula,followed by the roots.Furthermore,the MtC3H gene was found to respond to exogenous hormones such as gibberellin(GA),auxin(IAA),and methyl jasmonate(MeJA),with a particularly heightened sensitivity to MeJA induction.Subcellular localization analysis indicated that the MtC3H protein predominantly localized in the nucleus and cytoplasm.Notably,yeast expression analysis revealed that the MtC3H protein exhibited non-toxic properties and possessed transcriptional self-activation activity.This study provides a reference for further exploring the role of MtC3H gene.关键词
