肝细胞癌组织中细胞周期蛋白25C及其mRNA表达观察OACSTPCD
Expression changes of cell division cycle 25C protein and mRNA in hepatocellular carcinoma tissues
目的 观察肝细胞癌组织中细胞周期蛋白25C及其 mRNA的表达变化.方法 40例经病理检查明确诊断为肝细胞癌的患者,均行肿瘤切除术,术中留取肝细胞癌及癌旁组织,分别采用免疫组织化学法和原位杂交法检测肝细胞癌及其癌旁组织中CDC25C 蛋白和mRNA.结果 HCC组织和癌旁组织CDC25C 蛋白表达量分别为0.392±0.075、0.099±0.043,二者相比,P<0.05.HCC组织和癌旁组织CDC25C mRNA表达量分别为0.122±0.014、0.062±0.017,二者相比,P<0.05.与≤2级者相比,肿瘤分级>2级HCC患者HCC组织CDC25C 蛋白及mRNA表达量高(P均<0.05);与未有肝硬化者相比,伴有肝硬化的HCC患者HCC组织CDC25C 蛋白及mRN表达量高(P均<0.05).与男性相比,女性患者HCC组织CDC25C 蛋白表达量高(P<0.05).结论 肝细胞癌组织中CDC25C 蛋白和mRNA均高表达,CDC25C可能促进了肝细胞癌的发生发展.
Objective To observe the expression changes of cell division cycle 25C protein and mRNA in hepatocel-lular carcinoma tissues.Methods Forty patients with hepatocellular carcinoma confirmed by pathological examination underwent surgery,and hepatocellular carcinoma tissues and their adjacent tissues were collected during surgery.The ex-pression of CDC25C protein and mRNA in hepatocellular carcinoma and adjacent tissues were detected by in situ hybridiza-tion and immunohistochemistry,respectively.Results The relative expression levels of CDC25C protein in hepatocellu-lar carcinoma tissues and adjacent tissues were 0.392±0.075 and 0.099±0.043,respectively,with a significant differ-ence between them(P<0.05).The relative expression levels of CDC25C mRNA in hepatocellular carcinoma tissues and adjacent tissues were 0.122±0.014 and 0.062±0.017,respectively,with a significant difference between them(P<0.05).Compared with patients with grade≤2,patients with grade>2 had higher expression levels of CDC25C protein and mRNA in hepatocellular carcinoma tissues(both P<0.05).Compared with patients without liver cirrhosis,patients with liver cirrhosis had higher expression levels of CDC25C protein and mRNA in hepatocellular carcinoma tissues(both P<0.05).The expression of CDC25C protein in hepatocellular carcinoma tissues was higher in females than in males(P<0.05).Conclusion CDC25C protein and mRNA are highly expressed in hepatocellular carcinoma tissues,and CDC25C may promote the development and progression of hepatocellular carcinoma.
