目的 观察非小细胞肺癌(NSCLC)组织长链非编码核糖核酸(LncRNA)-WT1-AS、LncRNA-PRRT3-AS1的表达变化,探讨其对患者预后的预测效能.方法 选择病理检查明确诊断的NSCLC患者158例,均行肿瘤切除术,术中留取NSCLC癌组织与癌旁组织,采用实时荧光定量PCR法检测LncRNA-WT1-AS、LncRNA-PRRT3-AS1,分析LncRNA-WT1-AS、LncRNA-PRRT3-AS1与临床病理参数的关系,采用Kaplan-Meier法绘制LncRNA-WT1-AS、LncRNA-PRRT3-AS1的高表达和低表达NSCLC患者3年生存曲线,采用多因素COX回归分析法分析NSCLC患者预后不良的影响因素,绘制受试者工作特征曲线分析 LncRNA-WT1-AS、LncRNA-PRRT3-AS1表达对NSCLC预后不良的预测效能.结果 与癌旁组织相比,癌组织LncRNA-WT1-AS相对表达量低、LncRNA-PRRT3-AS1相对表达量高(P均<0.05).肿瘤直径≥3 cm、低分化程度、TNM分期Ⅱ期、淋巴结转移少的NSCLC患者LncRNA WT1-AS高表达比例低(χ2分别为7.680、4.300、13.830、4.135,P均<0.05);肿瘤直径≥3 cm、低分化程度、TNM分期Ⅱ期、淋巴结转移多的NSCLC患者LncRNA-PRRT3-AS1高表达比例高(χ2值分别为8.072、3.898、14.833、10.040,P均<0.05).与LncRNA-WT1-AS低表达者相比,LncRNA-WT1-AS高表达者3年生存率高(Log-rank=9.191,P<0.05);与LncRNA-PRRT3-AS1高表达者相比,LncRNA-PRRT3-AS1低表达者3年生存率高(Log-rank=8.783,P<0.053).肿瘤直径≥3cm、低分化程度、淋巴结转移、TNM分期Ⅱ期、LncRNA-WT1-AS低表达、LncRNA-PRRT3-AS1高表达是NSCLC患者预后不良的独立危险因素(P均<0.05).LncRNA-WT1-AS、LncRNA-PRRT3-AS1预测NSCLC预后不良的曲线下面积分别为0.767(95%CI:0.714~0.820)、0.848(95%CI:0.807~0.890),Cut-off值分别为2.810、2.705,敏感度分别为 53.8%、72.8%,特异度分别为 99.4%、84.8%.结论 NSCLC组织LncRNA-WT1-AS低表达、LncRNA-PRRT3-AS1 高表达.LncRNA-WT1-AS低表达、LncRNA-PRRT3-AS1 高表达的NSCLC患者预后较差,LncRNA-WT1-AS低表达和LncRNA-PRRT3-AS1高表达预测NSCLC预后不良的灵敏度及特异度均较高.

Objective To observe the expression changes of long non-coding RNA(LncRNA)-WT1-AS and Ln-cRNA-PRRT3-AS1 in non-small-cell lung cancer(NSCLC)tissues,and to explore their predictive efficacy for prognosis.Methods A total of 158 patients with NSCLC confirmed by pathological examination were selected.All patients under-went tumor resection.During the operation,NSCLC cancer tissues and adjacent tissues were collected.LncRNA-WT1-AS and LncRNA-PRRT3-AS1 were detected by real-time fluorescence quantitative PCR.The relationships between LncRNA-WT1-AS,LncRNA-PRRT3-AS1 and clinicopathological parameters were analyzed.Kaplan-Meier method was used to draw the 3-year survival curves of NSCLC patients with high and low expression of LncRNA-WT1-AS and LncRNA-PRRT3-AS1.Multivariate COX regression analysis was used to analyze the influencing factors of poor prognosis of NSCLC pa-tients.The receiver operating characteristic(ROC)curve was drawn to analyze the predictive efficacy of LncRNA-WT1-AS and LncRNA-PRRT3-AS1 expression on the poor prognosis of NSCLC.Results Compared with the adjacent tissues,the relative expression of LncRNA-WT1-AS was lower,and the relative expression of LncRNA-PRRT3-AS1 was higher in the cancer tissues(both P<0.05).The proportion of high expression of LncRNA WT1-AS in NSCLC patients with tumor diameter≥3 cm,poor differentiation,TNM stage Ⅱ,and less lymph node metastasis was low(χ2=7.680,4.300,13.830,and 4.135,respectively;P<0.05);the proportion of high expression of LncRNA-PRRT3-AS1 was higher in NSCLC patients with tumor diameter≥3cm,poor differentiation,TNM stage Ⅱ,and more lymph node metastasis was high(χ2=8.072,3.898,14.833,and 10.040,respectively;P<0.05).Compared with those with low expression of Ln-cRNA-WT1-AS,those with high expression of LncRNA-WT1-AS had a higher 3-year survival rate(Log-rank=9.191,P<0.05).Compared with those with high expression of LncRNA-PRRT3-AS1,those with low expression of LncRNA-PRRT3-AS1 had a higher 3-year survival rate(Log-rank=8.783,P<0.053).Tumor diameter≥3 cm,poor differentiation,lymph node metastasis,TNM stage Ⅱ,low expression of LncRNA-WT1-AS and high expression of LncRNA-PRRT3-AS1 were independent risk factors for poor prognosis of NSCLC patients(all P<0.05).The area under the curve of LncRNA-WT1-AS and LncRNA-PRRT3-AS1 in predicting poor prognosis of NSCLC was 0.767(95%CI:0.714-0.820)and 0.848(95%CI:0.807-0.890),respectively;the cut-off values were 2.810 and 2.705,the sensitivities were 53.8%and 72.8%,and the specificities were 99.4%and 84.8%,respectively.Conclusions LncRNA-WT1-AS is low expressed and LncRNA-PRRT3-AS1 is highly expressed in NSCLC tissues.The prognosis of NSCLC patients with low expression of LncRNA-WT1-AS and high expression of LncRNA-PRRT3-AS1 is poor.The sensitivity and specificity of low expression of LncRNA-WT1-AS and high expression of LncRNA-PRRT3-AS1 in predicting poor prognosis of NSCLC are high.
