Optimization of enzyme-producing medium of Aspergillus oryzae and effect of its solid-state fermentation products on feed intake and rumen pH in sheep

Abstract

The experiment aimed to explore the solid-state fermentation conditions affecting the production of Aspergillus oryzae xylanase,cellulase,and protease,as well as the impact of Aspergillus oryzae solid-state fermentation products on the rumen fermentation of sheep.A single-factor experiment was used to screen for the optimal ratio of carbon sources,nitrogen sources,and inorganic salts in the enzyme-producing medium.The cellulase,xylanase,and neutral protease activities in Aspergillus oryzae solid-state fermentation were measured.The results showed that the optimal carbon source,nitrogen source,and inorganic salts for enzyme production in Aspergillus oryzae solid-state fermentation were wheat bran,soybean meal,K2HPO4,and CaCl2,respectively.The optimal fermentation conditions were:80%wheat bran,20%soybean meal,0.5%K2HPO4,0.5%CaCl2,1%spore suspension inoculation,pH under natural conditions,and cultivation at 30℃for 5~7 days.Under these conditions,the cellulase activity in Aspergillus oryzae solid-state fermentation was 115.46 U/g,xylanase activity was 55.28 U/g,and neutral protease activity was 3 245.3 U/g.Twelve Kazakh sheep were selected and randomly divided into three groups,with four replicates per group,and each replicate consisted of one sheep.The control group was fed a basic diet,while the test Ⅰ group and test Ⅱ group were fed capsules containing 0.2%and 0.4%DM of Aspergillus oryzae fermentation products,respectively.The trial period was seven days.The results showed that the average daily weight gain,average daily feed intake,and average rumen pH value of the sheep in each test group were all higher than those in the control group.The study indicates that adjusting the types of nitrogen sources,carbon sources,and inorganic salts in the fermentation medium can enhance the enzyme production activity of Aspergillus oryzae solid-state fermentation,and adding 0.4%DM Aspergillus oryzae fermentation products to the diet is beneficial for increasing the average daily feed intake and average daily weight gain of sheep,as well as regulating the rumen pH value.
