目的 探究钙周期素结合蛋白(CacyBP)在食管癌患者中的表达情况,并检测CacyBP对食管癌TE-1细胞增殖、侵袭及迁移的影响及作用机制.方法 收集30例新鲜食管癌及癌旁正常组织,采用实时荧光定量PCR(qRT-PCR)及Western blot分别检测两组CacyBP mRNA及蛋白表达差异.同时收集90例患者临床病理学资料和食管癌及癌旁正常组织蜡块,免疫组化检测CacyBP表达情况,并分析CacyBP表达与临床病理学特征之间的相关性.人源食管鳞状细胞癌TE-1细胞分为空白对照组(control组)、转染空载体组(si-NC组)、转染siRNA CacyBP干扰片段组(si-CacyBP组),qRT-PCR验证转染效率.克隆形成实验、Transwell实验及划痕愈合实验分别检测TE-1细胞增殖、侵袭及迁移能力变化,Western blot检测上皮间质转化(EMT)相关分子(E-Cadherin、N-Cadherin、Vimentin、Snail和Slug)蛋白表达量情况.结果 食管癌组织中CacyBP表达量明显比癌旁正常组织高(P<0.05),且CacyBP的表达与肿瘤直径、淋巴结转移有关(P<0.05).与control组及si-NC组相比,si-CacyBP组TE-1细胞增殖、侵袭及迁移能力均明显降低(P<0.05),E-Cadherin蛋白表达量明显增高(P<0.05),N-Cadherin、Vimentin、Snail和Slug蛋白表达量明显降低(P<0.05).结论 CacyBP在食管癌中表达显著升高,敲低CacyBP表达抑制TE-1细胞增殖、侵袭及迁移,这可能与CacyBP调控EMT通路蛋白变化相关.

Objective To investigate the expression of calcyclin binding protein(CacyBP)in patients with esophageal cancer,and ex-plore the effect and mechanism of CacyBP on the proliferation,invasion and migration of esophageal cancer TE-1 cells.Methods Thirty cases of fresh esophageal cancer and adjacent normal tissues were collected to detect CacyBP mRNA and protein expression by real-time quantitative fluorescent PCR(qRT-PCR)and Western blot,respectively.At the same time,the clinical pathological data of 90 patients and paraffin blocks of esophageal cancer and adjacent normal tissues were collected to detect the expression of CacyBP by immunohistochemistry,and analyze the correlation between CacyBP expression and clinical pathological characteristics.Human esophageal squamous cell carcinoma TE-1 cells were divided into blank control group(control group),empty vector transfection group(si-NC group),and siRNA CacyBP interference fragment transfection group(si-CacyBP group),and qRT-PCR was used to verify the transfection efficiency.The proliferation,invasion and migration abilities of TE-1 cells were detected by clone formation assay,Tran-swell assay and scratch healing assay,respectively.The protein expression levels of EMT-related molecules(E-Cadherin,N-Cad-herin,Vimentin,Snail and Slug)were detected by Western blot.Results The expression of CacyBP in esophageal cancer tissues was significantly higher than that in adjacent normal tissues(P<0.05),and the expression of CacyBP was correlated with tumor dia-meter and lymph node metastasis(P<0.05).Compared with control group and si-NC group,the proliferation,invasion and migration abilities of TE-1 cells were significantly decreased in si-CacyBP group(P<0.05),and the expression level of E-Cadherin protein was significantly increased(P<0.05),and the expressions of N-Cadherin,Vimentin,Snail and Slug proteins were significantly decreased(P<0.05).Conclusion The expression of CacyBP is significantly increased in esophageal cancer.The knockdown of CacyBP expres-sion could inhibit the proliferation,invasion and migration abilities of TE-1 cells by regulating EMT pathway proteins.
