摘要

Abstract

Objective To observe the effect of electroacupuncture(EA)at Jiaji points(EX-B2)on expressions of silent information regulator 1(SIRT1)in nucleus pulposus cells of intervertebral disc(IVD)degeneration(IDD)model rabbits.Methods Totally 25 male New Zealand rabbits were randomly divided into normal group,model group,sham-operation group,model+EA group,and model+sham-EA group,5 in each group.Except the normal group,the rabbit models were prepared by longitudinal axial pressure to L4-L5 IDD rabbits for 28 days from the modeling device.And the rabbits of sham-operation group were only fitted with the modeling device without axial pressure.After completing modeling,the rabbits in the normal group,the model group,and the sham-model group were normally fed.EA at bilateral L4,L5 Jiaji points(EX-B2)were performed to rabbits in the model+EA group.EA alone was performed to rabbits in model+sham-EA group.The general condition and gait function scores of rabbits in each group were observed and recorded in each group.The signal intensities of the IVD were observed by magnetic resonance imaging(MRI)and the Pfirrmann classification performed.And the expression of SIRT1 in myeloid cells was detected by Western Blot and immunohistochemical assay.Results The rabbits in normal group were generally favorable with normal gait function.MRI scans showed a uniform bright high signal,with abundant expression of SIRT1 protein and higher rate of SIRT1-positive cells.The condition of the sham-model group was close to that of the normal group(P>0.05).Compared with the normal group,rabbits in the model group were in poor general conditions.Gait function scores significantly decreased(P<0.05),signal intensities of IVD disappeared,and the grading of Pfirrmann classification significantly increased(P<0.05),the expression of SIRT1 decreased(P<0.05),and the rate of SIRT1-positive cells decreased(P<0.05).Compared with the model group,the general conditions of rabbits in the model+EA group were improved.The mental conditions were better,gait function scores significantly increased(P<0.05),the signal intensities of IVD were significantly enhanced,and the grading of Pfirrmann classification lowered(P<0.05),the expression of SIRT1 increased(P<0.05),and the rate of SIRT1-positive cells increased(P<0.05).Some improvement showed in the model+sham-EA group,but the level of improvement was lower.Conclusion The axial pressure induced IDD in rabbits,while EA at Jiaji point(EX-B2)promoted the expression of SIRT1 in rabbit nucleus pulposus cells and delayed cellular aging to some extent,thus inhibiting IDD.
