为探究基于色差仪的CIBLAB量化方法在大豆种皮颜色遗传多样性分析中的应用潜力,本研究以274份大豆核心种质作为关联群体,基于CIBLAB方法,利用色差仪进行种皮颜色测定,获得6个颜色参数.利用主成分分析,明确表征大豆种皮颜色的最佳参数.在此基础上,通过遗传多样性和全基因组关联分析,挖掘与大豆种皮颜色显著关联的SNP位点.主成分分析表明,大豆种皮颜色主要包括黄、绿、黑和其他等4个类型,其中黄色占比较大,且黄蓝色度值(b*)是表征大豆种皮颜色的最佳参数.遗传多样性分析表明,黄色相关性状在品种间表现出丰富的遗传变异,变异系数为-35.53%~48.19%.对172份黄色种皮大豆全基因组关联分析,共检测到462个与b*显著相关的SNP位点,包括287个新位点和175个与已报道的I和L1位点重合的位点.综上,利用色差仪精准量化种皮颜色是一种能够客观评估大豆种皮颜色的新方法.利用该方法,检测到一些与大豆种皮颜色显著关联的新SNP位点,有助于进一步解析调控大豆种皮颜色的遗传机制.

Seed coat color is a crucial phenotypic feature in plants,whose accurate quantification is vital for variety classification,genetic analysis and agricultural applications.Using 274 soybean core germplasm as an association population,seed coat color was measured with a colorimeter based on CIBLAB to obtain six color parameters.Principal component analysis was used to identify the best parameters for characterizing soybean seed coat color.Based on this,genetic diversity and genome-wide association analysis were conducted to identify SNP loci significantly associated with soybean seed coat color.Soybean seed coat color was classified into four groups,including yellow,green,black,and other group.Principal component analysis indicated that yellow was the predominant color,and the yellow-blue value(b*)was the best parameter for characterizing soybean seed coat color.Genetic diversity analysis showed that yellow-related traits exhibited rich genetic variations among genotypes,with a coefficient of variation ranging from-35.53%to 48.19%.Genome-wide association analysis using 172 soybean genotypes with yellow seed coat color detected a total of 462 SNP loci significantly associated with the b*parameter,including 287 novel loci and 175 loci that coincide with the previously reported I and L1 loci known to control soybean seed coat color.Precise quantification of seed coat color using the colorimeter is a new method that could objectively verify soybean seed coat color.Using this method,several new SNP loci significantly associated with soybean seed coat color were detected,which would help to further elucidate the genetic mechanisms regulating soybean seed coat color.
