TIFY是一类植物特有的转录因子,参与植物的生长发育和逆境胁迫反应.目前,已在拟南芥和水稻等植物中报道了一些参与逆境胁迫的TIFY基因,但大豆中相关研究仍较少.为了明确大豆TIFY基因家族的蛋白质理化性质、基因结构、启动子顺式作用元件、基因进化以及表达特征,对其进行生物信息学分析,并进行转录组数据分析及干旱和盐胁迫qRT-PCR验证.结果显示:获得了 38个大豆TIFY家族基因,分布于16条染色体上,分为4个亚组,有10对片段重复基因和1对串联重复基因.GmTIFY蛋白均为亲水性蛋白,平均氨基酸序列长度为280.9 aa.启动子预测结果揭示GmTIFY基因启动子序列中富含光信号与激素和胁迫响应相关的顺式作用元件,表明TIFY家族基因可能参与大豆的生长发育调节过程.组织表达模式分析结果表明,GmTIFY在多个组织中表达差异显著,大多数基因在所有组织中的表达水平相对较低,甚至不表达.通过转录组数据分析及qRT-PCR实验发现,Glyma.01g204400、Glyma.05g235500、Glyma.09g077500、Glyma.13g112000、Glyma.15g179600 和 Glyma.15g184900 基因在干旱和盐胁迫下均上调表达,这些结果为深入研究大豆TIFY家族基因功能提供了参考.

TIFY is a class of plant-specific transcription factors involved in various plant growth and development processes and stress responses.At present,some TIFY genes involved in stress have been reported in Arabidopsis thaliana and rice,but there are still few related studies in soybean.In order to clarify the protein physical and chemical properties,gene structure,promoter cis-acting elements,gene evolution and expression characteristics of soybean TIFY gene family,this study performed bioinformatics analysis,transcriptome data analysis and qRT-PCR verification under drought and salt stress.The result showed:38 soybean TIFY family genes were obtained by bioinformatics method,which were distributed on 16 chromosomes and divided into four subgroups,including 10 pairs of fragment repeat genes and one pair of tandem repeat genes.The results of further analysis showed that all GmTIFY proteins were hydrophilic proteins and the average amino acid sequence length was 280.9 aa.Promoter prediction results showed that there are abundant light signals,hormones and stress in the promoter sequence of GmTIFY genes response-related cis-acting elements.The results of tissue expression pattern analysis showed that there were significant differences in the expression of GmTIFY in many tissues,and the expression level of most genes in all tissues was relatively low or even non-expressed.Through transcriptome data analysis and qRT-PCR experimental verification,it was found that Glyma.01g204400,Glyma.05g235500,Glyma.09g077500,Glyma.13g112000,Glyma.15g179600 and Glyma.15g184900 genes were upregulated under drought and salt stress.These results provide a reference for in-depth research on the functions of soybean TIFY family genes.
