基于心肌细胞衰老研究EGCG对铁死亡的影响
Effects of EGCG on ferroptosis based on H9C2 senescence

Abstract
Objective:To observe the effect of EGCG on cardiomyocyte senescence and ferroptosis.Methods:A senescence model of cardiomyocyte H9C2 was constructed using different concentrations of D-gal,and the proliferative activity of the cells was detected under the action of different concentrations of the drug for different times;different concentrations of EGCG was used to pretreat H9C2 and then carry out the construction of a senescence model,to detect the proliferation activity of the cells under the action of different concentrations of the drug for different times;the metabolic changes of intracellular ferroptosis were illustrat-ed by the mitochondrial morphology observation;ferrous ion(Fe2+)content detection,and reactive oxygen species(ROS)detec-tion were used to illustrate the metabolic changes of iron death in the cell;and cellular senescence β-galactosidase staining,immu-nofluorescence,and Western Blot were used to detect cellular senescence proteins p53,p21,and the ferroptosis protective pro-teins SCL7A11 and GPX4.Results:The effects of D-gal on cardiomyocytes was concentration-dependent and time-dependent,and negatively correlated;EGCG concentration over 5 μmol/L was negatively correlated with the proliferative activity of senes-cent cardiomyocytes;ferroptosis-specific alterations of cellular mitochondria in senescent H9C2 appeared,and improved after the drug was used;the content of Fe2+and ROS in senescent H9C2 was elevated,and the content of both was drastically reduced af-ter the use of EGCG;the degree of cell senescence of cardiomyocytes.The deeper the senescence of cardiomyocytes,the higher the levels of p53 and p21 proteins,and the lower the levels of GPX4 and SCL7A11 proteins.The levels of p53 and p21 proteins decreased,and the levels of GPX4 and SCL7A11 proteins increased after pretreatment with EGCG,with the same fluorescence results.Conclusion:the degree of iron death was aggravated in senescent cardiomyocytes,and EGCG could inhibit the senescence of the cells as well as ferroptosis.关键词
