摘要

Abstract

Objective To identify the the molecular mechanism of Dipsaci Radix in the treatment of osteoporotic fracture based on network pharmacology.Methods The main active components and targets of Dipsaci Radix was screened in the Traditional Chinese Medicine System Pharmacology Platform(TCMSP),and a prediction for osteoporotic fracture targets in the GeneCards was performed.The common targets of Dipsaci Radix and osteoporotic fracture were got by Venn diagrams.Cytoscape was used to construct a Chinese medicine-ingredient-disease-target regulation network,then the protein-protein interaction network was constructed and analyzed by STRING database.Finally,gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed by Bioconductor platform and R lan-guage.Results The main components of Dipsaci Radix for the treatment of osteoporotic fracture were 5 active components,such as β-sitosterol,Gentisin and Sylvestroside Ⅲ_qt,the targets for therapeutic effects might include 28 key targets such as caspase 3(CASP3),JUN,prostaglandin-endoperoxide synthase 2(PTGS2).Key pathways for therapeutic use included estrogen signaling pathway,host-pathogen interaction of human coronaviruses-apoptosis signaling pathway,interleukin-17 signaling pathway,corticotropin-releasing hormone signaling pathway and cellular response to organic cyclic compound sig-naling pathway.Conclusion This study shows the action characteristic of Dipsaci Radix in the treatment of osteoporotic fracture may be achieved by multi-components,multi-targets and multi-biological signaling pathways through network pharmacology,and it provide a scientific theoretical basis for the further research and experiments.
