基于大肠杆菌表达系统高效生产外源蛋白的自诱导培养基的研发OA北大核心CSTPCD
Development of self-induction medium for efficient production of recombinant protein based on Escherichia coli expression system
为提高大肠杆菌生产抗原的产量,实现企业降本增效,加快口蹄疫多表位疫苗的产业化进程,本研究拟研发适合大肠杆菌表达系统高效表达重组蛋白的自诱导培养基,并以实验室构建的口蹄疫多表位重组表达质粒为试验对象,通过正交试验、优化诱导温度,筛选和确定最佳自诱导培养基配伍.结果显示,在16 ℃诱导培养,自诱导培养基表达可溶性蛋白的产量为普通LB培养基的6倍,而成本却降低了 78%.这说明我们成功研发出了适合原核表达系统的自诱导培养基,其良好的成本效益性价比将有助于生物制品生产企业降低成本,提高产品的竞争力,具有广阔应用前景的产品.
The self-induction medium suitable for the expression system of Escherichia coli to ef-ficiently express recombinant protein was developed to increase the production of antigens,reduce costs and increase efficiency for enterprises.This study will be developed self-induction medium suitable for Escherichia coli expression system to express recombinant protein efficiently.The recom-binant expression plasmid with the gene of multi-epitope for foot-and-mouth disease antigen developed previously in our laboratory was used to verification of self-inducing medium.An optimal self-induc-ing culture medium was developed through orthogonal experiment and suitable induction temperature.The results showed that the yield of foot-and-mouth disease antigen produced by highly efficient self-in-duced medium was 6 times that of ordinary LB medium,and the cost of producing 1 mg foot-and-mouth dis-ease antigen was reduced by 78%.This indicated that we have successfully developed a self-induction medium suitable for prokaryotic expression systems.The good cost-effectiveness will help biological products enterprises reduce costs,improve product competitiveness,and create products with broad ap-plication prospects.
