褪黑素体内缓解急性铅中毒小鼠卵母细胞体外成熟的研究OA北大核心CSTPCD
Melatonin alleviates in vitro maturation of mice oocytes with acute lead poisoning
为探究注射褪黑素对急性铅中毒小鼠卵母细胞体外成熟的影响,以6~8周龄体况相近的ICR小鼠卵母细胞为研究对象,腹腔注射不同剂量的褪黑素(20、30、40 mg/kg体重),以第一极体排出作为卵母细胞成熟的标志,筛选出褪黑素适宜注射剂量继续进行后续试验.通过活性氧水平、ATP含量、线粒体分布以及细胞凋亡检测,分析注射褪黑素对急性铅中毒小鼠卵母细胞体外成熟的影响.结果显示,腹腔注射40 mg/kg的褪黑素可显著降低卵母细胞内活性氧水平、卵母细胞线粒体异常分布比率以及早期卵母细胞凋亡率,显著提高卵母细胞第一极体排出率及ATP含量.结果表明,腹腔注射40 mg/kg褪黑素可以提高急性铅中毒小鼠卵母细胞体外成熟的效果.
In order to investigate the effect of in vivo injection of melatonin on the maturation of oocytes in mice with acute lead poisoning,the oocytes of ICR mice aged 6-8 weeks with similar body con-dition were intraperitoneally injected with different doses of melatonin(20,30,40 mg/kg weight)in vivo,and the release of the first polar body was used as a sign of oocyte maturation.The optimal injection concentration of melatonin was determined,and the impact of melatonin injection on the in vitro matu-ration of mice oocytes with acute lead poisoning was analyzed by assessing reactive oxygen species,ATP content,mitochondrial distribution and apoptosis.The results showed that intraperitoneal injection of 40 mg/kg melatonin significantly reduced the reactive oxygen species in oocytes,the abnormal dis-tribution ratio of mitochondria,and the early apoptosis rate of oocytes.Additionally,it and signifi-cantly increased the first polar body excretion rate and ATP content of oocytes.In conclusion,in-traperitoneal injection of 40 mg/kg melatonin enhances the effect of oocyte maturation in vitro in mice with acute lead poisoning.
