Effect mechanism of dihydromyricetin on the tumor biological characteristics of endometrial cancer cells

目的 探讨二氢杨梅素对子宫内膜癌(EC)细胞增殖、凋亡及侵袭的影响,并分析其可能的作用机制.方法 取对数生长期Ishikawa细胞,分为对照组、20 µmol/L二氢杨梅素干预组、40 µmol/L二氢杨梅素干预组、80 µmol/L二氢杨梅素干预组,分别用不同终浓度的二氢杨梅素(0 µmol/L、20 µmol/L、40 µmol/L、80 µmol/L)处理Ishikawa细胞.CCK-8法和流式细胞术检测二氢杨梅素对细胞增殖和凋亡的影响.Transwell实验检测二氢杨梅素对细胞迁移、侵袭的影响.qRT-PCR和Western blot检测二氢杨梅素对细胞miR-21和PTEN表达的影响.结果 与对照组比较,二氢杨梅素干预组细胞增殖抑制率和凋亡率明显升高(P<0.05),且随着二氢杨梅素浓度的增加而逐渐升高(P<0.05);与对照组比较,二氢杨梅素干预组迁移及侵袭细胞数明显减少(P<0.05),且随着二氢杨梅素浓度的增加而逐渐减少(P<0.05);与对照组比较,二氢杨梅素干预组细胞miR-21表达明显降低(P<0.05),且随着二氢杨梅素浓度的增加而逐渐降低(P<0.05),PTEN mRNA和蛋白的表达水平明显增高(P<0.05),且随着二氢杨梅素浓度的增加而逐渐升高(P<0.05).结论 二氢杨梅素能够抑制EC细胞的生长及转移,其抑制作用与浓度呈正相关,其机制可能与二氢杨梅素对EC细胞miR-21/PTEN信号通路的影响有关.

Objective To explore the effects of dihydromyricetin on the proliferation,apoptosis,and invasion of endometrial cancer(EC)cells and its possible mechanisms.Methods Ishikawa cells in the logarithmic growth phase were taken and divided into the control group,20 µmol/L dihydromyricetin intervention group,40 µmol/L dihydromyricetin intervention group,and 80 µmol/L dihydromyricetin intervention group,which were treated with different final concentrations of dihydromyricetin(0 µmol/L,20 µmol/L,40 µmol/L,80 µmol/L).Then,CCK-8 assay and flow cytometry were used to detect the effects of dihydromyricetin on the cell proliferation and apoptosis.Transwell experiment was used to detect the effect of dihydromyricetin on the cell invasion.qRT-PCR and Western blot were used to detect the effects of dihydromyricetin on the cell expression of miR-21 and PTEN.Results Compared with the control group,the cell proliferation inhibition rate and apoptosis rate in the dihydromyricetin intervention group were significantly increased(P<0.05),and gradually increased with the increase of dihydromyricetin concentration(P<0.05).Compared with the control group,the numbers of migration and invasion cells in the dihydromyricetin intervention group were significantly decreased(P<0.05),and gradually decreased with the increase of dihydromyricetin concentration(P<0.05).Compared with the control group,the cell expression of miR-21 in the dihydromyricetin intervention group was significantly decreased(P<0.05),and gradually decreased with the increase of dihydromyricetin concentration(P<0.05),the expression levels of PTEN mRNA and protein were significantly increased(P<0.05),and gradually increased with the increase of dihydromyricetin concentration(P<0.05).Conclusion Dihydromyricetin can inhibit the growth and metastasis of EC cells,and the inhibitory effect is positively correlated with its concentration.The mechanism may be related to the effect of dihydromyricetin on the miR-21/PTEN signaling pathway of EC cells.
