Effects of dietary energy-protein level on rumen fermentation parameters and microflora of Tan sheep lambs
试验旨在探究饲粮能蛋水平对滩羊羔羊瘤胃发酵参数和微生物区系的影响.选择出生日期相近、体重(5.00±0.46)kg、健康的滩羊羔羊公羔60只,随机分为4个,每组3个重复,每个重复5只羔羊,分别为高能高蛋组即HEHP组(13.45 MJ/kg,20.55%)、高能低蛋组即HELP组(13.26 MJ/kg,16.10%)、低能高蛋组即LEHP组(11.40 MJ/kg,20.50%)和低能低蛋组即LELP组(11.50 MJ/kg,16.00%).预试期7d,正式试验期53 d.结果显示,LEHP组末重和平均日增重显著高于其他组(P<0.05).LEHP组的pH值最高,极显著高于HEHP组(P<0.01).HEHP组的瘤胃氨态氮含量最高,极显著高于其他组(P<0.01).HEHP组的微生物蛋白含量最高,极显著高于LELP组和HELP组(P<0.01),显著高于LEHP组(P<0.05);HEHP组的乙酸含量极显著高于其他组(P<0.01);HEHP组的丙酸含量最高,极显著高于LEHP组和HELP组(P<0.01),显著高于LELP组(P<0.05);LELP组的丁酸含量最高,显著高于HELP组(P<0.05).LELP组OTU数目、Ace指数、Chao1指数和Shannon指数最高,显著高于HEHP组(P<0.05).各试验组均以厚壁菌门、拟杆菌门、变形菌门为优势菌门,相对丰度均超过了 88%.HEHP组和LELP组瘤胃液中蓝藻门细菌相对丰度高于HELP组和LEHP组,LELP组的瘤胃液中的螺旋体门细菌相对丰度高于其他组.各试验组都以琥珀酸弧菌科UCG-001、普雷沃氏菌属和Shuttleworthia为优势菌属,HEHP组和HELP组的Shuttleworthia属相对丰度高于LEHP组和LELP组,HEHP组瘤胃液中小杆菌属相对丰度高于LEHP组.研究表明,滩羊羔羊早期补饲饲喂低能高蛋饲粮对瘤胃发酵参数和微生物区系效果较好.
The purpose of this experiment was to investigate the effect of dietary energy-protein level on rumen fermentation parameters and microflora of Tan sheep lambs.Sixty healthy Tan sheep lambs with similar birth dates and body weight of(5.00±0.46)kg were randomly divided into four groups with three replicates per group and five lambs per replicate.High energy high protein group,namely HEHP group(13.45 MJ/kg,20.55%).High energy low protein group,namely HELP group(13.26 MJ/kg,16.10%).Low energy high protein group,namely LEHP group(11.40 MJ/kg,20.50%).Low energy low protein group,namely LELP group(11.50 MJ/kg,16.00%).The pre-test lasted for seven days and the formal test lasted for 53 days.The results showed that the final weight and average daily gain of the LEHP group were significantly higher than those of the other groups(P<0.05).The pH value of the LEHP group was the highest,extremely higher than that of the HEHP group(P<0.01).The rumen ammonia nitrogen content of the HEHP group was the highest,extremely higher than the other three groups(P<0.01).The microbial protein content of the HEHP group was the highest,extremely higher than the LELP group and HELP group(P<0.01),and significantly higher than the LEHP group(P<0.05).The acetic acid content of the HEHP group was extremely higher than that of the other groups(P<0.01).The propionic acid content of the HEHP group was the highest,extremely higher than the LEHP group and HELP group(P<0.01),and significantly higher than the LELP group(P<0.05).The butyric acid content of the LELP group was the highest,significantly higher than the HELP group(P<0.05).The LELP group had the highest OTU number,Ace index,Chao1 index,and Shannon index,significantly higher than the HEHP group(P<0.05).All experimental groups were dominated by Firmicutes,Bacteroidetes,and Proteobacteria,with relative abundances all exceeding 88%.The relative abundance of Cyanobacteria in the rumen fluid of the HEHP group and LELP group was significantly higher than that of the HELP group and LEHP group(P<0.05),and the relative abundance of Spirochaetes in the rumen fluid of the LELP group was extremely higher than that of the other three groups(P<0.01).All experimental groups were dominated by Succinivibrio dextrinosolvens UCG-001,Prevotella,and Shuttleworthia,with the relative abundance of Shuttleworthia in the HEHP group and HELP group significantly higher than that of the LEHP group and LELP group(P<0.05),and the relative abundance of Eubacterium in the rumen fluid of the HEHP group was significantly higher than that of the LEHP group(P<0.05).The results show that early supplemental feeding of low energy high protein diet has a good effect on rumen fermentation parameters and microflora.
