试验旨在研究不同水平的蝗虫抗菌肽对仔猪生长性能、免疫功能及肠道屏障的影响.研究选择21日龄、体重约为7.0 kg的断奶仔猪,按照单因素试验设计分成4组,每组4个重复,每个重复8头断奶仔猪.各组仔猪分别饲喂添加0(对照组)、100、(A组)、200(B组)和400 mg/kg(C组)蝗虫抗菌肽的饲粮.预试期1 w,正式试验期6w.结果显示,与对照组相比,B组和C组仔猪的试验末重、平均 日增重均显著增加(P<0.05),所有抗菌肽组仔猪的腹泻率和料重比均降低(P<0.05).B组仔猪血清中二胺氧化酶(DAO)活性和白细胞介素-1β(IL-1β)含量显著降低(P<0.05),B组和C组血清中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量显著降低(P<0.05),所有抗菌肽组仔猪血清中白细胞介素-10(IL-10)含量显著升高(P<0.05).B组和C组仔猪结肠黏膜中闭锁蛋白(Occludin)、闭锁小带蛋白-1(ZO-1)mRNA相对表达量显著提高(P<0.05),Toll样受体4(TLR4)mRNA相对表达量显著降低(P<0.05).研究表明,仔猪饲粮中添加适量的蝗虫抗菌肽可以提高生长性能,降低腹泻率,调节血清免疫细胞因子水平,增强肠道屏障功能,蝗虫抗菌肽的最适添加水平为200 mg/kg.

The study aimed to investigate the effects of different levels of grasshopper antimicrobial peptides on the growth performance,immune function,and intestinal barrier of piglets.Twenty-one-day-old weaned piglets with a body weight of about 7.0 kg were divided into four groups with four replicates per group and eight weaned piglets per replicate according to a single-factor design.Piglets in each group were fed diets supplemented with 0(control group),100(group A),200(group B)and 400 mg/kg(group C),respectively.The pre-test period was one week and the formal test period was six weeks.The results showed that compared with the control group,the final weight and average daily gain of piglets in groups B and C were significantly increased(P<0.05),and the diarrhea rate and feed to gain ratio of piglets in all antimicrobial peptide groups were significantly decreased(P<0.05).The diamine oxidase(DAO)activity and interleukin-1β(IL-1β)content in serum of piglets in group B were significantly decreased(P<0.05),the concentrations of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in serum of groups B and C were significantly decreased(P<0.05),and the content of interleukin-10(IL-10)in serum of all antimicrobial peptide groups was significantly increased(P<0.05).The mRNA relative expression levels of atretic protein(Occludin)and atretic zonule protein-1(ZO-1)in colon mucosa of piglets in groups B and C were significantly increased(P<0.05),and the mRNA relative expression levels of toll-like receptor 4(TLR4)was significantly decreased(P<0.05).The study indicates that dietary supplementation of grasshopper antimicrobial peptides can increase growth performance,reduce diarrhea rate,regulate serum immune cytokine levels,and enhance intestinal barrier function of piglets,and the optimal supplemental level of grasshopper antimicrobial peptides is 200 mg/kg.
