精液冷冻保存是长期保护雄性生育力、转基因系、濒危物种种质资源的一个重要手段.冷冻保存的精液能跨越时空限制,有助于人工授精和体外受精等生殖技术快速发展.然而,冷冻-解冻过程会使精子遭受物理性、化学性以及氧化性损伤,导致冻融精子质量显著下降,影响其受精能力.其中,氧化性损伤主要表现为冷冻胁迫条件下精子的抗氧化防御系统被过量的活性氧(ROS)所淹没而形成的氧悖论状态,它被认为是冻融精子存活率及其受精能力下降的一个重要因素.目前,学者们正在尝试应用抗氧化剂来对抗过量ROS,从而平衡冷冻精子的抗氧化防御系统.本文主要综述了冷冻精子的氧化应激损伤、抗氧化剂的种类及其在精子冷冻保存中的应用现状,为深入研究抗氧化剂对冷冻精子的保护效应及其机制,以及进一步优化精液冷冻保存技术体系提供理论参考.

Semen cryopreservation is a crucial method for safeguarding male fertility,transgenic lines and germplasm resources of endangered species over extended periods.Cryopreserved semen can transcend the limitations of time and space,these advancements are beneficial for the rapid development of reproductive technologies such as artificial insemination and in vitro fertilization on a broad scale.However,the freeze-thawing process exerts detrimental effects on sperm,leading to its physical,chemical,and oxidative damages,which significantly reduce the quality of post-thawed sperm and affect the fertility.Among them,oxidative damage is mainly manifested as the oxygen paradox caused by the drowning of the antioxidant defense system of sperm by excessive production of reactive oxygen species(ROS)under freezing-thawing stress state.This phenomenon is considered as one of major impacts contributing to reduced survival rates and the insemination ability for post-thawed sperm.Currently,researchers are exploring the application of various antioxidants to counteract excess ROS,to balance the antioxidant defense system of frozen sperm cell.This paper mainly reviewed the recent research progress from three aspects:oxidative stress damage of sperm during the freezing process,types of antioxidants and their application status in cryopreserved sperm,so as to provide theoretical reference for further study the protective effects of antioxidants and their mechanism on frozen sperm,as well as further promoting the optimization of technical system on semen cryopreservation.
