GPX4基因多态性与幽门螺杆菌感染的关联研究OA北大核心CSTPCD
Association analysis of GPX4 gene polymorphisms with Helicobacter pylori infection
目的:分析谷胱甘肽过氧化酶4(GPX4)基因单核苷酸多态性(SNP)与幽门螺杆菌(H.pylori)感染的相关性.方法:选取506例研究样本,采用酶联免疫吸附试验与 14C尿素呼气试验检测H.pylori的感染情况,其中任意一种检测结果为阳性则判定为阳性;采用聚合酶链式反应-限制性片段长度多态性法检测GPX4 基因rs4807542、rs713041、rs2074451、rs3746162 的基因型分布;采用非条件性Logistic回归分析各SNP在H.pylori感染中的作用;采用SHEsis软件构建单体型,并分析其与H.pylori感染的关联性;采用广义多因子降维方法分析SNP-SNP的交互作用与H.pylori感染的关系.结果:GPX4 rs4807542、rs713041、rs2074451、rs3746162 在共显性、显性、隐性和超显性模型下均与H.pylori感染无关联(P>0.05);由rs4807542、rs713041、rs2074451、rs3746162构建的单体型未发现与H.pylori感染有相关性,这四个SNP在H.pylori感染中也不存在交互作用(P>0.05).结论:GPX4 rs4807542、rs713041、rs2074451、rs3746162可能在包头汉族人群H.pylori感染的过程中不起主要作用.
Objective:To analyze the association of single nucleotide polymorphisms(SNP)in glutathione peroxidase 4(GPX4)gene with Helicobacter pylori(H.pylori)infection.Methods:A total of 506 samples were selected to detect H.pylori in-fection by enzyme-linked immunosorbent assay(ELISA)and 14C Urea breath test(UBT),a positive result of either test was deter-mined as positive;GPX4 rs4807542,rs713041,rs713041 and rs3746162 were genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method;Unconditional logistic regression was used to analyze the association of each SNP with H.pylori infection.SHEsis software was used to constructed Haplotype to analyze its association with H.pylori in-fection.The relationship between SNP-SNP interaction and H.pylori infection was analyzed by generalized multi-factor dimen-sionality reduction method.Results:GPX4 rs4807542,rs713041,rs2074451 and rs3746162 were not associated with H.pylori in-fection in codominant,dominant,recessive,and overdominant models(P>0.05);Haplotypes constructed by rs4807542,rs713041,rs2074451 and rs3746162 were not founded to be associated with H.pylori infection,and these SNPs also did not have interaction in H.pylori infection(P>0.05).Conclusion:GPX4 rs4807542,rs713041,rs2074451 and rs3746162 may not play a major role in the process of H.pylori infection in Baotou Han population.
