枣DCL基因家族注释及其在枣疯病发生中的表达模式分析OA北大核心CSTPCD
Annotation of jujube DCL gene family and its expression pattern during jujube witches'broom disease
[目的]从枣基因组中注释枣 DCL(dicer-like)基因家族成员,对其在枣疯病发生过程中的表达模式进行分析,研究枣 DCL基因家族在响应植原体侵染中的作用.[方法]从 NCBI 网站下载冬枣基因组(Genebank 号JREP00000000)数据,以拟南芥 DCL蛋白序列为探针,使用生物信息学方法筛选得到枣 DCL 家族成员,分析其染色体位置、基因及蛋白结构、系统进化和保守基序特征,对枣 DCL 基因家族成员在枣疯病发生过程中的转录组数据进行分析,并对各成员在枣疯病发生过程中的动态进行实时定量反转录聚合酶链反应(qRT-PCR)定量.[结果]以 4 个拟南芥 DCL蛋白为探针,注释了 11 个枣 DCL成员(ZjDCLs),其蛋白分子质量范围为 15217.48~225623.49 D,等电点为 5.82~9.05.11 个枣 DCL基因不均匀分布在 6 条枣染色体上,其中 1 号染色体上的最多.根据和拟南芥 4 个 DCL 的进化关系可将 11 个枣 DCL 成员分为 4 类.基因结构分析表明,ZjDCL3 和ZjDCL7 外显子数量最多为 25 个,保守基序分析表明,枣 DCL 的结构较为保守.转录组数据分析表明,10 个枣DCL基因在枣疯病发生过程中有表达,ZjDCL6 在第Ⅳ个时期的表达量最高,qRT-PCR 定量分析表明,ZjDCL6,ZjDCL8 和 ZjDCL10 在不同时期健康树和病树之间差异变化较大.[结论]枣基因组中有 11 个 DCL 基因家族成员,结构较为保守,其中 ZjDCL6,ZjDCL8 和 ZjDCL10 在枣响应植原体侵染中起重要作用.
[Objective]The members of the jujube DCL gene family were annotated from the jujube ge-nome,and their expression patterns in the occurrence of jujube witches'broom disease were analyzed,and the role of the jujube DCL gene family in responding to phytoplasma infection was studied.[Method]The genome data of Jujube(Genebank No.JREP00000000)was downloaded from the NCBI website,and the DCL family members of Jujube were screened by bioinformatics methods using the DCL sequence of Arabidopsis thaliana as probe,and their chromosomal position,gene and protein struc-ture,phylogenetic and conserved motif characteristics were analyzed,the transcriptome data of the DCL gene family members of jujube during the progress of jujube witches'broom disease were analyzed,and the dynamics of each member in the process of jujube witches'broom disease were quantified by qRT-PCR.[Result]Eleven jujube DCLs(ZjDCLs)were annotated using four Arabidopsis DCL proteins as probes,with molecular weights ranging from 15217.48~225623.49 Da and isoelectric points ranging from 5.82~9.05.The 11 jujube DCL genes were unevenly distributed on 6 jujube chromosomes,most of which were on chromosome 1,and the 11 jujube DCLs could be divided into 4 categories according to the evolutionary relationship with the 4 DCLs in Arabidopsis,and the gene structure analysis showed that the number of ZjDCL3 and ZjDCL7 exons was up to 25,and the conserved motif analysis showed that the structure of jujube DCL was relatively conserved.Transcriptome data analysis showed that 10 jujube DCL genes were expressed during the process of jujube witches'broom disease,and the expre-ssion level of ZjDCL6 was the highest in the Ⅳ period,and qRT-PCR quantitative analysis showed that ZjDCL6,ZjDCL8 and ZjDCL10 were significantly different between healthy and diseased trees at diffe-rent periods.[Conclusion]There were 11 DCL gene family members in the jujube genome,and the structure was relatively conserved,among which ZjDCL6,ZjDCL8 and ZjDCL10 played an important role in the response of jujube to phytoplasma infection.
