蒸汽爆破忍冬藤饲料添加剂的益生功效研究OA北大核心CSTPCD
Study on the probiotic effect of steam exploded Lonicerae japonicae Caulis feed additive
[目的]探究蒸汽爆破忍冬藤(Lonicerae japonicae Caulis)的益生功效,为蒸汽爆破忍冬藤饲料添加剂的应用提供理论依据.[方法]以 4 种蒸汽爆破处理的忍冬藤(SELC-HOX、SELC-HCA、SELC-M1 和 SELC-M2)为研究对象,利用 1,1-二苯基-2-三硝基苯肼(DPPH)和 2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)法对比分析不同蒸汽爆破忍冬藤的自由基清除能力;借助体外模拟试验和 16S rRNA 基因扩增子测序技术探究不同蒸汽爆破忍冬藤对猪肠道微生物多样性、菌群结构的影响.[结果]与未蒸汽爆破忍冬藤相比,蒸汽爆破忍冬藤具有更强的自由基清除能力.体外模拟试验结果表明,蒸汽爆破忍冬藤能够显著降低猪肠道梭菌属 1(Clostridium_sen-su_stricto_1)、致病性肠埃希氏菌属-志贺氏菌属(Escherichia-Shigella)和柠檬酸杆菌属(Citrobacter)的相对丰度,显著提高乳杆菌属(Lactobacillus)相对丰度;相关性分析结果表明,蒸汽爆破忍冬藤的自由基清除能力及猪肠道乳杆菌属、链球菌属(Streptococcus)、肠球菌属(Enterococcus)、双歧杆菌属(Bifidobacterium)等益生菌属的相对丰度与低聚木糖含量显著正相关,梭菌属 1、致病性肠埃希氏菌属-志贺氏菌属和柠檬酸杆菌属相对丰度与低聚木糖含量显著负相关.[结论]蒸汽爆破忍冬藤具有更强的自由基清除能力和肠道微生物菌群调控功效,益生功效显著,可作为饲料添加剂用于畜牧养殖.
[Objective]Exploring the probiotic effect of steam exploded Lonicerae japonicae Caulis(LJC),and providing theoretical basis for the application of steam exploded LJC feed additives.[Method]Taking four steam exploded treatments(SELC-HOX,SELC-HCA,SELC-M1,and SELC-M2)of LJC as the research subjects,the study utilized the 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)methods to comparatively ana-lyze the free radical scavenging capacity of different steam exploded LJC.Additionally,the impact of different steam exploded LJC on the diversity and composition of pig intestinal microbiota was investi-gated through in vitro simulated experiments and 16S rRNA gene amplicon sequencing technology.[Result]Compared to untreated LJC,the steam exploded counterpart exhibited stronger free radical scavenging ability.The results of in vitro simulation experiments showed that steam explored LJC could significantly reduce the relative abundance of Clostridium_sensu_stricto_1,pathogenic Esche-richia-Shigella and Citrobacter,and significantly increased the relative abundance of lactobacillus in the pig intestine.The free radical scavenging ability of steam exploded LJC,as well as the relative abundance of probiotic genera such as Lactobacillus,Streptococcus,Eenterococcus and Bifidobacterium in the pig intestine,positively correlated with xylo-oligosaccharide content of LJC.While the relative abundance of Clostridium_sensu_stricto_1,pathogenic Escherichia-Shigella and Citrobacter negatively correlated with xylo-oligosaccharide content of LJC.[Conclusion]Steam exploded LJC(SELC-HOX),exhibits stronger free radical scavenging ability and modulation effects on gut microbiota com-pared to unexploded ones,the probiotic effect significant,suggesting its potential application as a feed additive in animal husbandry.
