中国紫杉状海门冬配子体和四分孢子体适宜温度和光照强度比较研究OA北大核心CSTPCD
A comparative study on the temperature and light intensity suitable for gametophytic and tetrasporophytic growth of Chinese Asparagopsis taxiformis
海门冬(Asparagopsis)作为一种反刍动物甲烷释放的有效抑制物,其生物量获取是促进相关应用领域持续发展的关键,因而海门冬人工栽培日益受到关注.本研究选取中国地区特有紫杉状海门冬配子体与四分孢子体为研究对象,光响应曲线测定结果显示配子体半饱合光强约为 56 μmol/(m2·s),四分孢子体的约为 88 μmol/(m2·s).光照强度对藻体光合作用及呼吸作用的影响结果显示,四分孢子体光合活性显著高于配子体,且具有较高的强光耐受能力.配子体和四分孢子体的光补偿点均约为 10 μmol/(m2·s),四分孢子体饱和光强在 250~270 μmol/(m2·s),相较于配子体的 100 μmol/(m2·s),显示出了更宽广的光强适应能力.在 15℃~31℃温度范围内,四分孢子体和配子体的光合作用系统均未受到显著影响,而四分孢子体有效量子产额在长时间 29℃条件下轻微下降.研究结果还显示,两种藻体在可溶性总蛋白、藻红蛋白及叶绿素a含量上未见显著差异,丝状四分孢子体因具有更大的比表面积,表现出显著高于配子体的光合效率.同时紫杉状海门冬四分孢子体具有较广的光强适应范围,因而是一种更适于基于反应器的规模化培养的藻体形态.
Research on the artificial cultivation of Asparagopsis has received increasing attention owing to its po-tential environmental benefits as an effective methane-release inhibitor for ruminants.Here,the light response curves of the gametophytes and tetrasporophytes of Asparagopsis taxiformis were determined first at the onset of light saturations(Ik)of 56 and 88 μmol/(m·s),respectively.Then,different light conditions were set up to study the effects of irradiation on the photosynthesis and respiration rates in the algae.The results showed that the tetraspo-rophytes exhibited a higher photosynthetic activity in comparison to gametophytes.The light compensation point,established using a liquid-phase oxygen electrode,was consistently identified at 10 μmol/(m·s)in both the game-tophytes and tetrasporophytes.Notably,the light intensity saturation point for the tetrasporophytes was 250-270 μmol/(m·s),substantially surpassing that of the gametophytes at 100 μmol/(m·s).However,the soluble total proteins,phycoerythrin,and chlorophyll-a contents did not differ significantly between the two generations.Within 15℃-31℃,the photosynthetic systems of both the tetrasporophytes and gametophytes did not exhibit any stress-related damage,while the effective quantum yield of the tetrasporophytes was slightly affected at>29℃during the fifth day of cultivation.These results also revealed no marked variations in the soluble total protein,phycoerythrin,and chlorophyll-a levels between the two algal morphologies.The filamentous tetrasporophyte ex-hibited remarkably higher photosynthetic efficiency than the gametophyte due to its larger surface area.Therefore,the tetrasporophyte of A.taxiformis is a prime candidate for future reactor-based large-scale commercial cultivation compared to the gametophyte due to its wide adaptability to varying light intensities and higher photosynthetic ac-tivity.
