摘要

Abstract

Amanita griseorosea is a lethal Amanita species within the sect.Phalloideae,mainly distributing in South China.In this study,toxins were detected through the methanol extraction and LC-MS analysis.Results showed that only α-amanitin was measured from this species,revealing an average concentration of(2 815.70±835.93)μg/g.In addition,genomic fragments from the MSDIN gene family were amplified by PCR for cloning and sequencing using degenerate primers.Seven MSDIN genes encoding different cyclic peptides were obtained.Sequence comparison and phylogenetic analysis indicated that one encodes the α-amanitin and another one encodes a cyclic peptide as previous reported.The remaining five all encode novel cyclic peptides.The above results thereafter provide an important foundation for further deep understanding of the toxicity and the capacity of cyclic peptide biosynthesis as well as the exploration of new cyclic peptides in A.griseorosea.
