口蹄疫是由口蹄疫病毒(foot-and-mouth disease virus,FMDV)感染偶蹄类动物(如猪、牛和羊)而引起的急性、高度传染性疾病.其非结构蛋白3Dpol在FMDV复制和致病过程中发挥着重要作用.为探究宿主蛋白与3Dpol蛋白的作用机制,本研究首先利用酵母双杂交和免疫共沉淀试验鉴定出VDAC1与FMDV 3Dpol相互作用,再运用过表达VDAC1和抑制剂试验发现VDAC1通过自噬途径抑制3Dpol蛋白的表达,最后发现下调VDAC1后对FMDV的复制有促进作用,表明VDAC1靶向3Dpol蛋白抑制FMDV复制.本研究揭示了宿主蛋白抑制FMDV复制的新机制,本研究为口蹄疫防控提供了新的方向和靶点.

foot-and-mouth disease is a highly contagious and severe illness triggered by the foot-and-mouth disease virus(FMDV)in even-toed ungulates like pigs,cattle,and sheep.The nonstructural protein 3Dpol of FMDV is crucial for the virus's replication and its role in causing the disease.In order to investigate the mechanism of host protein interaction with 3Dpol protein,this study firstly identi-fied the interaction between VDAC1 and FMDV 3Dpol using yeast two-hybrid and immunoprecipitation exper-iments,and then found the overexpression of VDAC1 and inhibitor experiments showed that VDAC1 inhibit-ed the expression of 3Dpol protein through autophagy pathway.Finally,it was found that down-regulation of VDAC1 had a promoting effect on FMDV replication,suggesting that VDAC1 was important in the replica-tion and pathogenicity of FMDV.This study reveals a new mechanism of host protein inhibition of FMDV replication,and the results provide a new direction and target for the prevention and control of FMD.
