摘要

Abstract

Objective:To explore the molecular mechanism of coptis rhizoma in the treatment of chronic periodontitis based on network pharmacology of traditional Chinese medicine.Methods:The effective active ingredients and targets of coptis rhizoma were screened using the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP),and the target genes of chronic periodontitis were screened using the genecards database.Then the intersection genes were obtain.The"drug-ingredient-target"network was constructed using Cytoscape.The protein-protein interaction(PPI)network,key subnetworks,and key genes were constructed using the String database.The key subnetworks were functionally enriched based on the kyoto encyclopedia of genes and genomes(KEGG)using the Webgestalt database.Results:The study screened out 14 effective ingredients of coptis rhizoma and 92 intersection genes,and constructed 2 key subnetworks,identifying the top 5 key targets:interleukin-6(IL-6),interleukin-1α(IL-1α),C-C motif chemokine 2(CCL2),interleukin-1β(IL-1β),interleukin-10(IL-10).KEGG results showed that the main enriched pathways included lactoferrin(LTF)danger signals,photodynamic therapy-induced nuclear factorκB(NF-κB),cytokines and inflammatory responses,adipokines,small cell lung cancer-related,peroxisome proliferator-activated receptor(PPAR)signaling pathways,etc.Conclusion:Coptis rhizoma can regulate chronic periodontitis by acting on targets such as IL-6,IL-1α,CCL2,IL-1β,IL-10,and regulating signaling pathways such as LTF danger signals,photodynamic therapy-induced NF-κB,cytokines and inflammatory responses,adipokines,small cell lung cancer-related,PPAR,etc.
