心肌细胞成熟障碍及其分子机制OACSTPCD
Impaired maturation of cardiomyocytes and its molecular mechanisms
心脏在胚胎晚期至成年期经历动态的成熟过程.但在心脏疾病或者心力衰竭时,心肌细胞重新激活胎儿时期基因而表现为形态和功能成熟障碍进而影响心脏功能.本文从成年型肌节和骨架蛋白下调、线粒体幼稚化,氧化磷酸化下降和增殖转录调控等方面对心肌成熟障碍的表现及其分子机制作总结,并通过调节能量底物和代谢过程、抑制进入细胞周期等探讨促心肌细胞成熟策略,为未来挖掘心力衰竭发病机制和改善措施提供理论依据.
The heart undergoes a dynamic maturation process from late embryonic stages to adulthood.However,in conditions such as cardiac disease or heart failure,cardiomyocytes tend to re-activate fetal gene expression,leading to morphological and functional maturation impairments that affect cardiac function.This review summarizes the manifesta-tions and molecular mechanisms of cardiomyocyte maturation impairment,focusing on the downregulation of adult sarcom-eric and cytoskeletal proteins,mitochondrial immaturity,reduced oxidative phosphorylation,and the dysregulation of pro-liferative transcriptional controls.Strategies to promote cardiomyocyte maturation,including regulation of energy substrates and metabolic processes,as well as inhibition of cell cycle re-entry,are discussed to provide theoretical support for un-derstanding heart failure mechanisms and improving treatment approaches.
