含挥发油饮片砂仁标准汤剂的质量评价方法研究OACSTPCD
Quality Evaluation of Standard Decoction of Amomi Fructus Containing Volatile Oil
目的:通过分析砂仁标准汤剂出膏率、乙酸龙脑酯及挥发油的含量和转移率、特征图谱,建立其质量评价方法.方法:按要求制备 15 批砂仁标准汤剂,测定出膏率、乙酸龙脑酯和挥发油含量,并计算转移率.同时采用CORTECS T3 色谱柱(100 mm×2.1 mm,1.6 μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,检测波长为 260 nm,流速为 0.35 mL·min-1,柱温为 30℃,建立标准汤剂特征图谱分析方法.结果:出膏率为 12.3%~17.7%,乙酸龙脑酯转移率为 3.9%~10.5%,挥发油转移率为 5.6%~15.6%.在最佳色谱条件下,采集到的砂仁标准汤剂特征图谱实现了对多种化学成分的展现,共标定 11 个特征峰,采用液相色谱-质谱法(LC-MS/MS)和对照品比对确认其中 7 个特征峰,其中峰 1 为原儿茶酸、峰 4 为香草酸、峰 5 为儿茶素、峰 7 为原花青素B2、峰8 为表儿茶素、峰 10 为异槲皮苷、峰 11 为槲皮苷,采用"中药色谱指纹图谱相似度评价系统"(2012 版),得到 15批砂仁标准汤剂的相似度为 0.984~0.999.聚为两类,2 个主成分的累积方差贡献率为 81.579%.结论:建立的含量测定及特征图谱方法适用性与准确度好,能较全面地反映砂仁标准汤剂的内在质量,可为砂仁配方颗粒的制备提供参考.
Objective:To establish a quality evaluation method for the standard decoction of Amomi Fructus decoction pieces based on the extract yield,content and transfer rates of index components,and characteristic chromatogram.Methods:15 batches of standard decoction were prepared according to the requirements.The extract yield and the content of bornyl acetate and volatile oil were determined,and the transfer rates were calculated.The chromatography was performed in a CORTECS T3 column(100 mm×2.1 mm,1.6 μm)with acetonitrile(A)-0.1%phosphoric acid aqueous solution(B)as the mobile phase by gradient elution at the flow rate of 0.35 mL·min-1 and the column temperature of 30℃.The detection wavelength was set at 260 nm.A characteristic chromatogram-based method for evaluating the standard decoction was thus established.Results:The extract yields,bornyl acetate transfer rates,and volatile oil transfer rates in 15 batches of Amomi Fructus samples varied within the ranges of 12.3%-17.7%,3.9%-10.5%,and 5.6%-15.6%,respectively.The characteristic chromatogram of Amomi Fructus standard decoction was collected under the optimal chromatographic conditions,revealing a variety of chemical components.A total of 11 characteristic peaks were marked,and 7 of them were identified by liquid chromatography(LC)-tandem mass spectrometry(MS/MS)and reference substances.Specifically,peaks 1,4,5,7,8,10,and 11 represented protocatechuic acid,vanillic acid,catechin,procyanidin B2,epicatechin,isoquercetin,and quercetin,respectively.According to the similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(2012 edition),the fingerprint similarity of the 15 batches of Amomi Fructus standard decoction was wihtin the range of 0.984-0.999.The cumulative variance contribution rate of the first two principal components was 81.579%.Conclusion:The method for content determination and the characteristic chromatogram established in this study had good applicability and accuracy.They can comprehensively reflect the intrinsic quality of standard decoction of Amomi Fructus decoction pieces and provide reference for producing the formula granules of Amomi Fructus.
