Abstract

To realize the utilization of traditional Chinese herbs post-harvest waste in aquaculture,the stems and leaves of Radix astragalus,Radix rhubarb and Radix bupleuri were used as the main fermentation raw materials,and Bacillus subtilis SA-2 isolated from the sediment of shrimp pond was used as the bacteria strain for fermentation.With the ratio of solid to liquid,inoculation amount,fermentation time,and fermentation temperature as variable factors,the amount of viable bacteria and the content of total polysaccharides after fermentation as evaluation indexes,the aerobic fermentation conditions for stems and leaves of compound Chinese herbs were optimized using an orthogonal test.The results showed that when the viable bacteria amount was taken as the evaluation index,the optimal culture conditions were as follows:Solid to liquid ratio 2∶1,inoculation amount of bacteria 2%,fermentation temperature 34℃,and fermentation time 72 h.Under these conditions,the viable bacteria amount reached 1.09×1010 CFU/mL.When the content of total polysaccharides after fermentation was taken as the evaluation index,the optimal culture conditions were as follows:Solid to liquid ratio 1∶2,inoculation amount of bacteria 6%,fermentation temperature 31℃,and the fermentation time was 72 h.Under these conditions,the total polysaccharide content could reach 38.3 mg/g,which was 14.1 times higher than that before fermentation.The study indicates that the stems and leaves of compound Chinese herbs fermented by Bacillus subtilis can be used as potential feed additive in aquaculture.
