草莓试管苗玻璃化现象探究及生理特性分析OA北大核心
Vitrification phenomenon exploration and physiological characteristics analysis of strawberry in vitro seedlings
为探究草莓试管苗玻璃化现象和生理特性,以'红颜''越秀''红珍珠''圣诞红''光点''白雪公主''妙香''章姬'这8个品种草莓试管苗为试材,分析不同品种草莓试管苗玻璃化差异;通过测定'红颜'草莓玻璃化试管苗的含水率、叶绿素相对含量、相对电导率、丙二醛含量和氧化酶活性等指标,进一步分析草莓玻璃化试管苗生理性状差异;测定不同外植体消毒液HgCl2的含量和处理时间对试管苗玻璃化的影响以及不同玻璃化程度的试管苗驯化移栽后的生理性状差异.结果表明:1)不同草莓品种玻璃化率不同,'白雪公主'草莓玻璃化率最高,为31.0%;'圣诞红'草莓玻璃化率最低,为14.0%.2)玻璃化草莓试管苗的含水率、相对电导率和丙二醛含量等显著高于正常草莓试管苗,株高和叶绿素相对含量显著低于正常草莓试管苗(P<0.05).3)玻璃化草莓试管苗诱导过氧化氢酶(CAT)活性显著降低,过氧化物酶(POD)活性显著升高(P<0.05).4)HgCl2含量越高,消毒时间越长,玻璃化率越高,使用0.5%HgCl2处理16 min时,'红颜'草莓玻璃化率达到38.2%.5)重度玻璃化草莓试管苗驯化移栽后,株高、叶绿素相对含量、茎粗和干鲜重等均显著低于正常草莓生根苗(P<0.05).综上,草莓试管苗的玻璃化现象可能是一种氧化胁迫反应.
In order to investigate the vitrification phenomenon and physiological characteristics of strawberry test-tube seedlings,eight test-tube seedling varieties,'Benihoppe','Yuexiu','Red Pearl','Ssanta','Light Spot','Snow White','Miaoxiang'and'Akihime'were taken as test materials to analyze the vitrification difference of different varieties of strawberry test-tube seedlings.By measuring the moisture content,relative content of chlorophyll,relative conductivity,malondialdehyde content and oxidase activity of the vitrification test tube seedling of'Benihoppe'strawberry,the physiological characteristics of the vitrification test tube seedling were further analyzed.In addition,the effects of content and treatment time of HgCl2 on the vitrification of test-tube seedlings and the differences of physiological traits of test-tube seedlings with different degrees of vitrification after acclimation and transplanting were investigated after domestication and transplanting.The results showed that:1)Different strawberry varieties have different vitrification rates.Among these,the'Snow White'strawberry had the highest vitrification rate of 31.0%,while the'Ssanta'strawberry had the lowest vitrification rate of 14.0%.2)The water content,relative electrical conductivity and malondialdehyde content of vitrification strawberry test-tube seedlings were significantly higher than those of normal strawberry test-tube seedlings,while the plant height and chlorophyll content were significantly lower than those of normal strawberry test-tube seedlings(P<0.05).3)The induction catalase(CAT)activity of vitrification strawberry tube seedlings decreased significantly,and the activity of peroxidase(POD)increased significantly(P<0.05).4)The higher the content of HgCl2,the longer the disinfection time,and the higher the vitrification rate.After treatment with 0.5%HgCl2 for 16 min,the vitrification rate of'Benihoppe'strawberry reached 38.2%.5)After the domestication and transplanting of severe vitrification strawberry test tube seedlings,the plant height,relative chlorophyll content,thick stem,dry and fresh weight were significantly lower than those of normal strawberry root seedlings(P<0.05).In conclusion,the vitrification of strawberry test tube seedlings may be an oxidative stress reaction.
