百合花药水通道蛋白LoPIP2;6基因克隆、亚细胞定位与表达OA北大核心
Cloning,subcellular localization and expression analysis of aquaporin LoPIP2;6 gene of lily anther
为进一步探究水通道蛋白PIP2亚家族成员在花药发育及开裂过程中发挥的作用,以百合花药为试验材料,利用同源克隆法获得LoPIP2;6基因,采用DNAMAN、MEGA软件对蛋白结构和系统进化树进行分析,利用烟草瞬时表达检测亚细胞定位,通过实时荧光定量PCR检测基因表达.结果表明:LoPIP2;6基因开放阅读框有849 bp,编码282个氨基酸,为疏水性蛋白,含有6个典型的跨膜螺旋结构域及2个NPA基序,与小兰屿蝴蝶兰(Phalaenopsis equestris)、铁皮石斛(Dendrobium catenatum)的亲缘关系最近;LoPIP2;6蛋白亚细胞定位于质膜和内质网上;LoPIP2;6基因在花药发育处于四分体时期表达量最高,其次为独立小孢子时期.综上,推测LoPIP2;6可能在花药发育前期细胞增长、花药发育后期药室隔膜与裂口细胞的水分转移过程中发挥作用,研究结果为进一步探究水通道蛋白调控百合花药发育及开裂提供了理论参考.
In order to further explore the role of members of the PIP2 subfamily of aquaporins in anther development and dehiscence,lily anthers were taken as experimental materials.The LoPIP2;6 gene was obtained from lily anthers using homologous cloning method.Protein structure and evolutionary tree analysis were conducted using DNAMAN and MEGA software.Subcellular localization was detected using tobacco transient expression,and gene expression was detected using real-time fluorescence quantitative PCR.The results were as follows:The open reading frame of LoPIP2;6 gene contained 849 bp encoding 282 amino acids.The LoPIP2;6 was a hydrophobic protein that contained 6 typical transmembrane helical domains and 2 NPA motifs.LoPIP2;6 protein had closer relationship with Phalaenopsis equestris and Dendrobium catenatum,and the protein was localized to the plasma membrane and endoplasmic reticulum.The LoPIP2;6 gene displayed the highest expression level at the tetrad stage,followed by the independent microspore stage.It was speculated that LoPIP2;6 might play a role in the cell growth during the early stage of anther development,as well as in the water transfer of anther connective tissue and crack zone cells during the late stage.In conclusion,this study provides a theoretical reference for further exploring the regulation of lily anther development and dehiscence by aquaporins.
