Accumulation Characteristics and Key Gene Expression Analysis Involved in Anthocyanin Biosynthesis in Camellia japonica

Abstract
[Objective]This study aims to investigate the accumulation of metabolites and the expression characteristics of key genes in camellia(C.)japonica flowers with varying flower colors,as well as to analyze the relationship between the two factors.[Method]Six different color traits of C.japonica petals were selected as materials to determine the contents of anthocyanins,total phenols,flavonoids,and proanthocyanidins.Real-time fluorescence quantitative PCR was employed to assess the expression levels of genes associated with product synthesis,and a network diagram illustrating the relationship between metabolite accumulation and gene expression was constructed.[Result]The accumulation of anthocyanins in dark colored cultivars of C.japonica were found to be higher than that in light and white cultivars.Notably,the purple-red cultivar'Midnight'exhibited significantly higher contents of anthocyanins,flavonoids,and proanthocyanidins,reaching 572.18 µg/g,18.93 mg/g,and 97.29 mg/g,respectively.Furthermore,the total phenolic content in dark and light colored cultivars exceeded that of moderately colored cultivars,with the red cultivar'Chidan'displaying the highest content at 103.13 mg/g,while'Midnight'also showed a relative high level of accumulation.Quantitative expression analysis revealed that gene expression levels varied specifically among different cultivars.'Midnight'demonstrated high expression of upstream genes(CHS and F3H)and low expression of downstream genes(F3'H,DFR,ANS,and UFGT).Correlation analysis indicated that flavonoid and proanthocyanidin accumulation was positively correlated with the expression of F3H,ANS,and UFGT.Additionally,anthocyanin accumulation was positively correlated with the expression of CHS,CHI,and F3'H,and negatively correlated with F3H,DFR,and ANS.[Conclusion]The C.japonica'Midnight'exhibits a high accumulation of anthocyanins,total phenols,flavonoids,and proanthocyanidins.The upstream and downstream genes in the synthesis pathway have both promoting and inhibitory effects on anthocyanin synthesis and accumulation,respectively,while downstream genes generally promote the accumulation of flavonoids and proanthocyanidins.关键词
