Effects of Converting Eucalypt Plantations to Dendrocalamus latiflorus Forests on Soil and Root Microbial Communities Based on Metagenomic Sequencing

Abstract

To reveal changes in microbial communities of soils and fine roots after conversion of eucalypt plan-tations to Dendrocalamus latiflorus forests and to provide theoretical basis for sustainable management of D.lat-iflorus forests,taking D.latiflorus forests and eucalypt plantations located in drinking water source areas of sub-urbs of Nanning as research objects,chemical properties in fine roots of D.latiflorus and eucalypt and in soils of D.latiflorus forests and eucalypt plantations as well as carbon(C),nitrogen(N),and phosphorus(P)stoichio-metric ratios were analyzed.Using metagenomic sequencing technology,structures and relative abundances of microbial communities were analyzed in soils of D.latiflorus forests and eucalypt plantations and in fine roots of D.latiflorus and eucalypt.Results showed that after converting eucalypt plantations to D.latiflorus forests,total carbon content,C∶N and C∶P in fine roots of D.latiflorus were lower than those in fine roots of eucalypt,while contents of total phosphorus,total nitrogen,total potassium and N∶P in fine roots of D.latiflorus were higher than those in fine roots of eucalypt.pH,contents of total carbon and hydrolyzable nitrogen as well as C∶N,C∶P and N∶P in soils of D.latiflorus forests were higher than those in soils of eucalypt plantations,while contents of total phosphorus,total nitrogen,total potassium and available phosphorus in soils of D.latiflorus forests were lower than those in soils of eucalypt plantations.Dominant microbial phyla in soils of D.latiflorus forests and eucalypt plantations and in fine roots of D.latiflorus and eucalypt were Proteobacteria,Actinobacteria and Ac-idobacteria.Relative abundances of Actinobacteria and Proteobacteria had significant correlations with soil pH and total phosphorus content,while relative abundance of Acidobacteria had significant correlations with soil pH,C∶N and hydrolyzable nitrogen content.In fine roots of eucalypt and D.latiflorus,relative abundance of Ac-tinobacteria had significantly negative correlations with total carbon content and C∶N,relative abundance of As-comycetes had significantly positive correlation with C∶N,and relative abundance of Acidobacteria had signifi-cantly negative correlation with total potassium content.Redundancy analysis showed that environmental fac-tors that had stronger influences on microbial community structures were contents of total carbon and total phos-phorus and soil pH.When managing D.latiflorus pure forests,attention should be paid to inputs of carbon,ni-trogen,and phosphorus as well as to regulation of soil pH.
