Study on the role of HSP90 functional antibody on stemness characteristics of gastric cancer stem cells

目的:研究热休克蛋白90(HSP90)功能性抗体对胃癌干细胞干性特征的作用,并初步探讨相关机制.方法:无血清悬浮培养胃癌HGC27细胞获得细胞球细胞(Sphere细胞),HSP90功能性抗体9B8处理Sphere细胞,CCK-8法检测9B8对胃癌干细胞耐药、增殖的作用,Trans-well小室检测9B8对胃癌干细胞迁移、侵袭的作用,甲基纤维素成球检测9B8对胃癌干细胞自我更新的作用,流式细胞术检测9B8对胃癌干细胞ALDH、CD44表达的作用,免疫荧光检测HSP90在胃癌干细胞中的表达,Western blot检测胃癌干细胞中HSP90、干性相关蛋白(SOX2、OCT4)、耐药相关蛋白(TOP2A、ABCG2)、上皮-间质转化(EMT)相关蛋白(Vimentin、Snail、E-cadherin)及PI3K/AKT/mTOR信号通路相关蛋白表达.结果:胃癌干细胞中HSP90、ALDH、CD44 表达高于亲本细胞,且其增殖、耐药、迁移、侵袭及自我更新能力明显强于亲本细胞(P<0.01).HSP90功能性抗体9B8处理组与对照组相比,胃癌干细胞增殖没有差异(P>0.05),但迁移、侵袭、耐药和自我更新能力显著低于对照组(P<0.01),ALDH、CD44表达比例明显下降(P<0.01).HSP90功能性抗体处理组HSP90、ABCG2、TOP2A、Vimentin、Snail、PI3K、p-PI3K、AKT、p-AKT、mTOR、p-mTOR表达下降,E-cadherin表达增加.结论:HSP90功能性抗体9B8可能通过抑制AKT/PI3K/mTOR通路活性,影响胃癌干细胞相关干性基因/蛋白、耐药基因/蛋白、EMT相关基因/蛋白表达,进而影响胃癌干细胞自我更新、耐药、侵袭、迁移等干性特征,为胃癌干细胞靶向治疗提供了新的思路.

Objective:To investigate the role of the functional antibody heat shock protein 90(HSP90)on stemness character-istics of gastric cancer stem cells and to preliminarily explore the related mechanisms.Methods:Using serum-free suspension cul-ture of gastric cancer HGC27 cells to obtain cytosphere cells(Sphere cells);treating Sphere cells with HSP90 functional antibody 9B8 and using CCK-8 assay to detect the role of 9B8 on gastric cancer stem cell resistance and proliferation;using Trans-well chamber to detect the role of 9B8 on migration and invasion of gastric cancer stem cells;using methylcellulose sphere-forming as-say to detect the role of 9B8 on the self-renewal of gastric cancer stem cells;using flow cytometry to detect the role of 9B8 on the expression of ALDH and CD44 in gastric cancer stem cells;using immunofluorescence to detect the expression of HSP90 in gas-tric cancer stem cells,and Western blot to detect the expression of HSP90,stemness-associated proteins(SOX2 and OCT4),drug-resistance-associated proteins(TOP2A and ABCG2)in gastric cancer stem cells,and epithelial-mesenchymal transition(EMT)-related proteins(Vimentin,Snail,and E-cadherin),as well as the expression of proteins related to PI3K/AKT/mTOR signaling pathway.Results:The expression of HSP90,ALDH,and CD44 in gastric cancer stem cells was higher than that of pa-rental cells,and their proliferation,drug resistance,migration,invasion,and self-renewal abilities were significantly stronger than those of parental cells(P<0.01).There was no difference in the proliferation of gastric cancer stem cells in the HSP90 functional antibody between the 9B8-treated group and the control group(P>0.05),but the migration,invasion,drug resistance,and self-renewal abilities were significantly lower than those in the control group(P<0.01),and the proportion of ALDH and CD44 ex-pression was significantly decreased(P<0.01).The expressions of HSP90,ABCG2,TOP2A,Vimentin,Snail,PI3K,p-PI3K,AKT,p-AKT,mTOR,and p-mTOR were decreased in the HSP90 functional antibody-treated group,and the expression of E-cadherin was increased.Conclusion:HSP90 functional antibody 9B8 may affect the expression of gastric cancer stem cell-re-lated stemness genes/proteins,drug resistance genes/proteins,and EMT-related genes/proteins by inhibiting the activity of AKT/PI3K/mTOR pathway,which in turn affects the stemness characteristics of gastric cancer stem cells,such as self-renewal,drug resistance,invasion,and migration,and thus provides a new way of thinking for the targeting therapy of gastric cancer stem cells.
