目的:观察大鼠在干热环境力竭运动及给予氨基酸维生素后,大鼠骨骼肌组织病理形态学和骨骼肌损伤相关等指标,探讨氨基酸维生素对干热环境大鼠运动骨骼肌损伤的保护作用.方法:将36只SPF雄性SD大鼠随机分为空白对照组、常温运动组、干热运动组、低剂量组、中剂量组、高剂量组,运动结束后取大鼠比目鱼肌组织观察病理学变化.ELISA检测血清炎症因子指标MCP-1;氧化应激指标CAT、GSH-Px;qRT-PCR检测大鼠比目鱼肌Nrf2/HO-1/NQO1 mRNA相对表达量;WB检测大鼠比目鱼肌Nrf2、HO-1、NQO1蛋白表达水平.结果:(1)大鼠核心体温:力竭运动后较运动前升高(P<0.05),低、中、高剂量组核心体温较干热运动组下降(P<0.05);(2)大鼠力竭时间:干热运动组力竭时间较常温运动降低(P<0.05),低、中、高剂量组力竭时间较干热运动组升高(P<0.05);(3)炎症因子及氧化应激:干热运动组血清MCP-1较常温运动组升高(P<0.05),MCP-1中、高剂量组较干热运动组降低(P<0.05);干热运动组血清CAT和GSH-Px水平较常温运动组降低(P<0.05);CAT高剂量组较干热运动组升高(P<0.05),GSH-Px低、中、高剂量组较干热运动组升高(P<0.05);(4)qRT-PCR检测:干热运动组Nrf2、NQO1 mRNA相对表达水平显著较常温运动组降低(P<0.05),Nrf2、NQO1相对表达水平低、中、高剂量组较干热运动组升高(P<0.05),HO-1相对表达水平中、高剂量组较干热运动组升高(P<0.05);(5)WB检测:干热运动组Nrf2、HO-1蛋白表达量较常温运动组降低(P<0.05),Nrf2表达量低、中、高剂量组较干热运动组升高(P<0.05),HO-1表达量中、高剂量组较干热运动组升高(P<0.05),NQO1高剂量组较干热运动组升高(P<0.05).结论:干热环境可能会加重运动骨骼肌损伤,氨基酸维生素可能对干热环境大鼠运动骨骼肌损伤的严重程度发挥保护作用.

Objective:The histopathological morphology of skeletal muscle and the related indexes of skeletal muscle injury in rats were observed,and the protective effect of amino acid vitamins on sports skeletal muscle injury in rats in dry and hot environ-ment was investigated.Methods:Thirty-six SPF male SD rats were randomly divided into blank control group,room temperature exercise group,dry heat exercise group,low dose group,medium dose group and high dose group.ELISA was used to detect se-rum inflammatory factor MCP-1.oxidative stress indicators CAT,GSH-Px;The relative expression levels of nuclear factor ery-throid-2 related factor 2 and Nrf2HO-1NQO1 mRNA in rat soleus muscle were detected by qRT-PCR.WB was used to detect the protein expression levels of Nrf2,HO-1 and NQO1 in rat soleus.Results:(1)The core body temperature of rats increased after exhaustion exercise compared with that before exercise(P<0.05),and the core body temperature of the low,medium and high dose groups decreased compared with that of the dry heat exercise group(P<0.05);(2)Exhaustion time of rats:the exhaustion time of the dry heat exercise group was lower than that of normal temperature exercise(P<0.05),and the exhaustion time of low,medium and high dose groups decreased compared with that of normal temperature exercise(P<0.05).The exhaustion time of the high-dose group was higher than that of the dry-heat exercise group(P<0.05);(3)Inflammatory factors and oxidative stress:the serum MCP-1 of the dry-heat exercise group was higher than that of the room-temperature exercise group(P<0.05),the MCP-1 medium-dose and high-dose groups were lower than those of the dry-heat exercise group(P<0.05),and the serum levels of CAT and GSH-Px in the dry-heat exercise group were lower than those of the room-temperature exercise group(P<0.05).The high-dose CAT group was higher than that of the dry-heat exercise group(P<0.05),and the low,medium,and The mRNA expression levels of Nrf2 and NQO1 in the high-;dose exercise group were significantly lower than those in the room-tem-perature exercise group(P<0.05),the low-,medium-,and high-dose Nrf2 and NQO1 groups were higher than those in the dry-heat exercise group(P<0.05),and the HO-1 medium-and high-dose groups were higher than those in the dry-heat exercise group(P<0.05),and the HO-1 medium-and high-dose groups were higher than those in the dry-heat exercise group(P<0.05);(5)WB detection:Nrf2 and The expression of HO-1 protein decreased compared with the normal temperature exercise group(P<0.05),the expression of Nrf2 in the low,medium and high dose groups was higher than that in the dry heat exercise group(P<0.05),the expression of HO-1 in the medium and high dose groups was higher than that in the dry heat exercise group(P<0.05),and the NQO1 high dose group was higher than that in the dry heat exercise group(P<0.05).Conclusion:Dry heat environment may aggravate sports skeletal muscle injury,and amino acid vitamins can play a protective role in the severity of sports skeletal muscle injury in rats in dry heat environment.
