基于ROS/NLRP3介导的泪腺上皮细胞焦亡探讨梅花生津饮治疗干燥综合征的机制研究OA北大核心
Mechanism of Meihua Shengjin Decoction in the treatment of Sjögren's syndrome through ROS/NLRP3-mediated pyroptosis of lacrimal epithelial cells
目的:观察梅花生津饮对泪腺上皮细胞ROS/NLRP3信号通路的影响,从而阐明其通过细胞焦亡治疗干燥综合征的分子机制.方法:NOD小鼠给予梅花生津饮进行干预,并以C57BL/6J小鼠作为空白对照.采用H&E染色观察各组小鼠泪腺组织病理的改变,酚红棉线染色法检测泪液分泌,ELISA法检测氧化应激指标.在此基础上,以H2O2诱导泪腺上皮细胞构建细胞模型,并给予梅花生津饮含药血清进行干预.采用流式细胞检测细胞ROS和焦亡的水平,ELISA法检测乳酸脱氢酶的释放,采用Western blot检测NLRP3相关蛋白的表达.结果:与模型组相比,梅花生津饮能够显著增加干燥综合征小鼠泪液分泌,改善泪腺组织病理评分、降低血清MDA的含量和增加SOD的活性,差异具有统计学意义(P<0.01).此外,与模型组相比,梅花生津饮能够显著降低泪腺上皮细胞ROS、焦亡水平和LDH的释放,下调NLRP3蛋白的表达与降低Cleaved Caspase-1/Caspase-1、GSDMD-N/GSDMD-FL、Mature IL-1β/Pro-IL-1β和Mature IL-18/Pro-IL-18的比值,差异具有统计学意义(P<0.01).结论:梅花生津饮通过抑制ROS/NLRP3通路的激活,抑制泪腺上皮细胞的焦亡,从而抑制炎症反应,达到治疗干燥综合征的作用.
Objective:To observe the effect of Meihua Shengjin Decoction on the ROS/NLRP3 signaling pathway in lacrimal epithelial cells,thereby elucidating its molecular mechanism in treating Sjögren's syndrome through cell pyroptosis.Methods:NOD mice were administered with Meihua Shengjin Decoction for intervention,and C57BL/6J mice were used as the control.H&E staining was used to observe the histopathological changes in the lacrimal glands.Phenol red thread staining was employed to detect tear secretion,and ELISA was used to measure oxidative stress indicators.Additionally,cellular model was constructed by inducing lacrimal epithelial cells with H2O2,and the cells were treated with Meihua Shengjin Decoction-containing serum.Flow cy-tometry was used to detect cellular ROS and pyroptosis levels,ELISA was utilized to measure the release of lactate dehydroge-nase,and Western blot was performed to assess the expression of NLRP3-related proteins.Results:Compared to the model group,Meihua Shengjin Decoction significantly increased tear secretion in mice with Sjögren's Syndrome,improved the histopath-ological score of the lacrimal glands,reduced serum MDA content,and increased SOD activity,with statistically significant differ-ences(P<0.01).Furthermore,compared to the model group,Meihua Shengjin Decoction significantly reduced the levels of ROS and pyroptosis,and LDH release in lacrimal epithelial cells.It also downregulated the expression of NLRP3 protein and decreased the ratios of Cleaved Caspase-1/Caspase-1,GSDMD-N/GSDMD-FL,Mature IL-1β/Pro-IL-1β,and Mature IL-18/Pro-IL-18,with statistically significant differences(P<0.01).Conclusion:Meihua Shengjin decoction inhibits the activation of the ROS/NL-RP3 pathway,suppressing pyroptosis in lacrimal epithelial cells,thereby inhibiting inflammatory responses and treating Sjögren's syndrome.
